We developed a routine method for the determination of adriamycin (ADR) and its metabolites in the serum and plasma by use of high performance liquid chromatography (HPLC) with fluorescence detector and "protein-coated ODS" column. The samples, adjusted to about pH 3 by 1 M H8PO4, are injected directly onto the column and eluted stepwise with phosphate buffer and a mixture of the same buffer and acetonitrile. The chromatograms are monitored at Ex 470 nm and Em 585 nm. 127 samples were assayed continuously by use of only one column and an automated HPLC. The sera or plasmas of human and rabbit to which ADR was administered were analysed by the present method and the total fluorescence method. ADR, adriamycinol and minor metabolites, were detected on chromatograms. Both methods were compared each other and discussed. The present method is simpler and more economical than the other ones previously described.
