Obesity is a major public health problem among the industrial countries. The WHO estimates that over 300 million people are clinically obese. Obesity is associated with prothrombic and proinflammation states, hypertension, dyslipidemia, hyperglycemia, insulin resistance. These obesity-related abnormalities are commonly referred to as the metabolic syndrome. Recently, it was reported that adipocyte itself synthesizes and secrets a large variety of molecules called adipokine including leptin, adiponectin, resistin, TNF α , TGF β , and PAI-1
etc, which act both at local (autocrine/paracrine) and systemic (endocrine) levels. Excessive production and secretion of adipokines may cause the obesity-associated metabolic syndrome. In this review, we summarize our studies on gene regulations of plasminogen activator inhibitor-1 (PAI-1) by adipocyte-enriched transcription factor, PPAR γ and that of resistin with treatment of resveratrol in adipocytes and obese mice.
