The possible role of antibody as a selective pressure on antigenic mutants of poliovirus in nature was investigated in vitro. A mutant resistant to a monospecific antibody with a definite specificity was readily obtained by several cycles of neutralization of Mahoney strain with a monospecific antibody and multiplication in monkey kidney (MS) cells. Mutants resistant to more than two different monospecific antibodies were also readily obtained in a similar manner. Studies on the antigenicity of these mutants by kinetic neutralization tests revealed that the Mahoney strain underwent a progressive serological variation as it became successively resistant to one to five different monospecific antibodies isolated from anti-Mahoney serum.

A single administration (1 to 10 mg/kg) of rice dwarf virus RNA (RDV-RNA) prior to virus challenge reduced the mortality of mice infected with western equine encephalitis (WEE) virus. The protective effect of RDV-RNA was significantly higher than that of polyinosinic-polycytidylic acid. However, when these RNAs were given after virus infection, the protective effect was negligible. The titer of circulating interferon in mice reached a peak about 5 hr after injection of these RNAs and remained at this level for about 24 hr. Viremia in mice infected with a lethal dose of WEE virus was markedly suppressed by the treatment of mice with these RNAs. A pathological examination of mice treated with a lethal dose of RDV-RNA revealed marked changes including degeneration and karyorrhexis in the lymphoid tissues of the spleen.

Diminutive viral RNAs recovered from tobacco leaves inoculated with ³²P-TMV were investigated. At 3.5 hr after inoculation, most of the viral RNA without coat protein revealed two peaks after sucrose density gradient analysis of SDS-extract from 12, 000 × g leaf pellet. The first peak appeared between bacterial ribosomal RNA of 16 S and 5 S and the second peak was around 5 S. These two peaks were digestible with RNase and they appeared as early as 5 min after inoculation. These diminutive RNAs seemed to be derived from partially uncoated parental virus by abscission of their naked RNA tails. The active formation of these diminutive RNAs and their early appearance after inoculation seemed to indicate that most of the inoculated TMV received incomplete uncoating.
