We established an Na₂S-free, large-scale overexpression system of deriving CODH II from thermophilic bacterium Carboxydothermus hydrogenoformans in Escherichia coli using a large-scale fermentor. Recombinant-CODH II showed a CO oxidation activity of 9,600 U/mg. In addition, recombinant-CODH II exhibited considerable CO₂ reduction activity, of 16.9 U/mg.
