Thiamine (I) reacted with ethyl isocyanate to give three moles adduct (II) and four moles adduct (III). N-Ethylcarbamoylthiamine (VIII) reacted with ethyl isocyanate to afford a mixture of three moles adducts, and separation of these isomers revealed the formation of perhydrofurothiazole derivatives (IX, X). NaBH4 reduction of these diastereomers gave dihydro (XIII) and tetrahydro (XV) compounds, respectively.
