Instead of the currently used heated and solidified egg-media, in drug resistance test of tuberculous bacilli, the author, using blood agar penicillin media (modified Tarshis media), which is easily manufactured and highly sensitive in bacilli detection, togetherwith Ogawa's media as the control, conductedd the present study.
Resistances against streptomycin (SM), para-aminosalicylic acid (PAS) and isonicotinic acid hydrazid (INAH) were measured through direct method by using the TB patients' sputa. Resistances of the strains isolated from the sputa against these drugs were measured through indirect method. Pre-treatment of the sputa was done by tri-sodium phosphate method. Comparison was made in modification of drug's power-value in the media and extent of growth in both media for every resisting bacilli. SM's power-value in the media was measured physiologically, while that of PAS and INAH were measured by chemical measurement. Growth of resisting bacilli in both media was tested, by using human type TB bacilli (H37Rv strains), on its sensitive original strains and each of SM, PAS and INAH resistant variants. The results obtained are as follows;
1) As for resistance against dihydrostreptomycin, no marked difference was observed between the modified Tarshis media and Ogawa's media, containing a double volume of DHSM.
2) Resistance against PAS appeared with less extent in the modified Tarshis media than in Ogawa's media, containing the same amount of INAH.
3) Resistance against INAH appeared in higher extent in the modified T arshis media than in Ogawa's media, containing the same amount of INAH,
4) Practically, no change was observed in the drug power value of DHSM, PAS and INAH, in the modified Tarshis media. On the contrary, decreases of 43% in DHSM, 74% in PAS and 82% in INAH were observed in Ogawa's media.
5) In the test using human type H37Rv strains, it was observed that INAH resistant bacilli showed a better growth in the modified Tarshis media than in Ogawa's media, and SM resistant bacilli, compared with the sensitive bacilli, showed a better growth in the modified Tarshis media.
6) In the modified Tarshis media, the drug power-value is accurate, and the media is suited to the growth of resistant bacilli, (resistance against INAH can be observed at early stage). Moreover, the media is manufactured very easily, thus, it is highly suitable in the test of drug resistance of tuberculous bacilli.
