In order to develop an optimum program for the subcutaneous open-loop insulin infusion which can attain the physiological meal-related plasma insulin profile, the kinetics of subcutaneously administered insulin was studied by two-compartment analysis. An exponentially decreasing insulin infusion pattern was determined by mathematically deconvoluting an ideal insulin curve. Furthermore, a new insulin infusion system which cosisted of portable and preprogrammable infusion pump was developed to enable the abovementioned program. Thus, these new program and apparatus succeeded in obtaining an expected plasma insulin profile as well as glycemic normalization and proved to be feasible and promising for the long-term glycemic and insulin normalization.
