To progress developmental events normally, it is crucial to regulate temporal and spatial expression patterns of genes participate in these processes correctly.
We previously revealed that the Arabidopsis ERECTA gene, which encodes a receptor-like protein kinase, is involved in morphogenesis of aerial parts and expressed in the entire meristems and organ primordia. We also reported the cis-regulatory sequences responsible for ERECTA gene expression. A yeast one-hybrid screen for proteins that associate with this region led to the identification of ERBP1, a potential regulatory component of ERECTA transcription.
For a further investigation into the ERECTA transcriptional regulatory mechanism, now we are taking both genetic and biochemical approaches. First, to identify cis-regulatory elements, we have examined the 5' upstream regulatory region using linker scanning mutational analysis. Second, we have performed a yeast two-hybrid screen using ERBP1 as bait and isolated some candidates possible to involve ERECTA transcription as an interactor for ERBP1.
