The effects of heat and ethanol treatments used to sterilize cysts of potato cyst nematodes (Globodera pallida and G. rostochiensis) were determined. Cysts isolated from the soil were completely sterilized after soaking in water in 1.5-ml plastic tubes and heated at over 60°C for 1 h. Similarly, they were sterilized by soaking in 70% ethanol for over 1 h in the tube. The sterilization method using 70% ethanol is thought to be highly practical because it does not require special equipment. In addition, the sterilization of soils which are infested by cyst nematodes was tested as a possible treatment method. The cysts of G. rostochiensis in soil survived the heating conditions that were previously reported. However, all cysts of G. pallida and G. rostochiensis were completely sterilized when the soil was heated at 90°C for 1 day. These sterilization methods can be useful for minimizing the risks of nematode spread in Japan.
