The Mannich reaction of 4-methyl-2-pentanone in both acidic and basic media has been extensively studied. In the acidic medium, 4-methyl-2-pentanone reacts with morpholine and formaldehyde to give 1-N-morpholino-5-methyl-3-hexanone (4) and 1-N-morpholino-2-isopropyl-3-butanone (5), respectively, in good yield (81%, (4)/(5)=69/31). On the other hand, the reaction in basic medium afforded the same products ((4)/(5)=8/92) in 43% yields. The time-course variation of this reaction was also examined and the mechanism for the reaction is suggested. The structure of the products was elucidated by chemical evidences and spectroscopic data.
