Three kinds of lipases (Novo SP525, Amano PS, and Amano AK) were encapsulated in inorganic matrices by sol-gel method for synthesis of chiral compounds by kinetic resolution. The inorganic matrices were prepared by hydrolyzing silane precursor (vinyltriethoxysilane, VTES; methyltrimethoxcysilane, MTMS; tetramethoxysilane, TMS; or propyltrimethoxysilane, PTMS) in water with sodium fluoride as the catalyst, following by mixing lipase aqueous solution and gelling in room temperature for 65 h, grounding in a motor. Enantioselective hydrolysis of 2-octyl acetate by sol-gel lipase SP525 was done in the mixture of phosphate buffer (0.1 M, pH 7) and acetone. The sol-gel lipase prepared from VTES showed best hydrolysis activity and its reactivity is higher than that of sol-gel lipase prepared from the mixture of PTMS/TMS = 5/1 (molar ratio). The sol-gel lipase had good reactivity after incubating at 90oC for 1 h, however, the reactivity of free lipase was decreased markedly even at 60oC.
