Biopharmaceuticals, including nucleic acids and proteins, frequently require cold-chain logistics and are commonly administered via invasive injections. This study describes the development of dry powder inhalers using electrospinning (ES) technique to address the dual challenges of biopharmaceuticals. Nanofiber mats composed of polyvinyl alcohol and model therapeutics (α-chymotrypsin or plasmid DNA) were fabricated and subsequently processed into inhalable powders via freeze-milling. The ES powders successfully maintained high protein activity. Furthermore, in vivo studies demonstrated excellent aerosol performance and sustained pulmonary gene expression. Therefore, ES presents a promising, heat-free platform for non-invasive respiratory drug delivery.
