An assay method of vitamin B_<12> in serum and whole blood applicable to the Euglena method was studied. 1-3 ml of sample was digested with trypsin and takadiastase at 37° for 48 hr. After heating at 100° for 30 min. with a reaction of pH 5.0,0.1 ml of 10% metaphosphoric acid was added in cold to remove the minute amount of protein which might interfere with the turbidimetry. After centrifuging, the supernant liquid was adjusted to pH of assay media and diluted with water to the proper volume. The recoveries of added vitamin B_<12> were 93-106%. The mean values of eight healthy subjects were 0.334mμg/ml in whole blood and 0.335 mμg/ml in sera respectively.
