The particulate fraction R_<12>, which contains mainly respiratory granule, and supernatant fraction S_<12> from B. aneurinolyticus were investigated on thiaminase activity and some enzymatic activities concerning in the role of thiaminase in this bacterial cell. Respiratory activity of R_<12> was observed only with succinate and α-ketoglutarate as substrates, and the addition of S_<12> was needed for utilization of glutamate or pyrrolidone carboxylate. Respiratory activity of R_<12> were controlled by ATP. S_<12> had no respiratory activity, however it showed the remarkable dehydrogenase activity, while R_<12> and S_<12> inter-depended on glutamate metabolism. The effect of S_<12> on aerobic glutamate metabolism of R_<12> was partially replaced by hydroxymethylpyrimidine, one of the products by thiaminase II action. Thiaminase II activity was predominant in S_<12> compared with R_<12>, however some enzymatic activity remained in R_<12>. This remained activity of R_<12> did not removed by washing with water. Glutamate-alanine transaminase activity was observed in S_<12> when it was measured by the estimation of alanine, but the reverse reaction was not observed. This phenomenon may suggest that the produced glutamate is converted into ninhydrin-negative substance, and the decomposition of glutamate was accelerated by hydroxymethylpyrimidine.
