During the course of experiment on the nature of crystalline urease, bis-(p-nitrophenyl) disulfide (I) was found to be a potent inhibitor of urease activity. It was shown that (I) was more inhibitory at an alkaline medium of higher pH and the inhibition was progressive with time. The inhibition of urease was prevented or recovered by the addition of an excess amount of such sulfhydryl compounds as reduced glutathione and cysteine. Possible mechanism of inhibition by bis-(p-nitrophenyl) disulfide seems to be due to disulfide-exchange reactions between the inhibitor and sulfhydryl groups of urease with a progressive loss of its activity.
