A method for the determination of swertiamarin, the bitter component of the herb of Swertia japonica, was established by the use of high-speed liquid chromatography. Swertiamarin in Swertia Herba was separated on a 20 cm column of LiChrosorb RP-8, using the Shimadzu Du Pont LC-3A liquid chromatograph with 1.5% tetrahydrofuran as the desorption solution. Detection was achieved at 235 nm by a SPD-2A multiwave phothometric detector. The precision of this method was about ±1% and the detection limit was several ng (S/N ratio, 3). Swertiamarin in Swertiae Herba was extracted with water and the extract was injected directly into the column. The content of swertiamarin was calculated from the calibration curve of the previously prepared standard. The cycle time of analysis was about 15 min. This mathod is considered to be useful for the evaluation of Swertia Herba.
