By the addition of 20-30% H2O2 to alkaline copper solution of over pH 9.2, dark brown precipitate of CuO⋅H2O appears. The presence of α-amino acid group in the solution prevents the precipitation the solution remaining blue and clear. This reaction was, therefore, thought to be applicable for a qualitative estimation of α-amino acid group. With the exception of citric and saccharic acids, many other compounds do not show this effect. Detectable amount is 0.05×(1-5)mg. of amino acid (molecuear weight taken as 200).
