The effect of guanidine, hydrazine, tropolone, and 2-aminoethylisothiuronium (AET) derivatives on the incorporation of acetate-1-14C into cholesterol was examined in the rat liver homogenates. Incorporation of acetate-1-<14>C into cholesterol was inhibited to varying degrees by all of the test compounds. Among these compounds 1, 1'-decamethylenediguanidine dihydrochloride, carbonyl cyanide phenylhydrazone, phenylhydrazine hydrochloride, 5-nitrosotropolone, and 2-(2-aminoethyl)-1'(p-chlorophenyl) isothiuronium bromide hydrobromide showed potent inhibitory action. The inhibitory action of these compounds was not affected by adding ATP (10-5-10-3M) exogenously into the incubation system. This result suggests that these compounds have sites of action other than blocking the high energy-forming system in mitochondria.
