A method for the determination of furfurals (furfural and 5-hydroxymethylfurfural) in aged sake by gas chromatography was established. Five ml of sample was reacted with 4 ml of 2, 4-dinitro phenylhydrazine (2, 4-DNP) solution for 30 min at 80 °C. The solution was shaken with 30ml chloroform. The chloroform layer was concentrated to dryness in vacuo, and dissolved in 0.5 ml of 0.16% methyl-2-furfural-2, 4-DNP (internal Standard) in ethyl acetate. One μl of the solution was analyzed using a gas chromatograph (Shimadzu GC-14 A). This method will ensure determination of furfurals in aged sake with 104-108% recovery.
The results of analysis on 44 samples aged for 3-20 years were: furfural 2-4 ppm, and 5-hydroxymethylfurfural 1-289 ppm. Furfurals were considered to be one of the characteristic components of aged sake.
