The chloroplast NDH complex is involved in PSI cyclic and chlororespiratory electron transport in higher plants. We previously showed that NDH interacts with at least two copies of PSI to form the unique NDH-PSI supercomplex, which is required for protection of NDH from high-light stress. Despite the substantial progress that has been made in elucidating the structure of NDH, the mechanism of the NDH assembly has remained elusive. Here, we employed ten Arabidopsis mutants specifically defective in accumulation of the NDH subcomplex A to investigate the assembly process of this subcomplex. Our detailed proteomic, biochemical and genetic studies provide some insights into the mechanism of the NDH subcomplex A assembly and our working model will be discussed.
