It was reported previously that methyl linoleate hydroperoxide (ML-HPO) absorbed oxygen and the oxidized ML-HPO contained great quantities of methyl 8-formyl octanoate (MFO) and malonaldehyde (MA).
In this report, the oxidative products of ML-HPO were examined. A main product which had HO-, HOO- and trans -C=C- groups was separated from the oxidized ML-HPO by column chromatography. The molecular weight of the product was about 620 and decreased to a half by catalytic hydrogenation. It was indicated that the product was a dimer and the monomer units were combined by peroxy group (-O-O-). The dimer released easily MFO and MA on heating.
The reaction on glutathione and toxicity of the dimer were researched. Sulfhydryl group of glutathione decreased more quickly by the dimer than by ML-HPO when they were incubated with glutathione in phosphate buffer solution (pH 7.6) at 37°C. The dimer inhibited the growth of mice as well as ML-HPO.
