Electrokinetic potential of Sephadex G-10, -15, -25, -50, -75, -100, and -200 gel particles swollen with water was measured by the moving boundary electrophoretic method. Relationships were investigated between the ζ-potential and characteristics of swelling process in the xerogels, and elution parameters of several food dyes and peptides in gel filtration. It was found that the Sephadex gel particles have negative ζ-potential and its values increased in proportion to the increase of G-numbers, namely, water regain of the gels. This finding suggested that the values of ζ-potential was mainly related to the swelling degree of the gels. A different property between lower and higher G-number gels was shown on the basis of distinctive linear relationships between ζ-potential and swelling characteristics of the xerogels such as effective diffusion coefficient of water. Elution parameters observed in gel filtration of peptides increased relatively to the values of ζ-potential, while the those of several food dyes decreased through adsorption.
