    4p heterodimeric complex is well known as a transcriptional activator for the genes regulated by inositol and choline, such as the
    INO1
    gene. Apl2p is a large subunit of the yeast adaptin complex, an adaptor complex required for the clathrin coat to bind to the membrane. We found that
    Ino
    2p,
    Ino
    4p, and Apl2p form a ternary complex. This interaction was initially observed in a yeast two-hybrid study and subsequently verified by co-immunoprecipitation.
    Ino
    2p and
    Ino
    4p bind to Apl2p in the same region of Apl2p, viz., at the middle part and the C-terminal part.
    Ino
    2p and
    Ino
    4p bind to Apl2p independently, but more strongly when both are present. Furthermore, a disruption of APL2 together with
    INO2
    or
    INO4
    rendered yeast cells sensitive to oxidative stress.
    INO
    2-APL2
    double disruptants also showed growth inability in non-fermentable carbon sources, such as glycerol. These results indicate a genetic interaction between APL2,

    Relapsed acute lymphoblastic leukemia (ALL) has a poor prognosis. Inotuzumab Ozogamicin (

    InO
    ) is a novel therapeutic drug for the treatment of relapsed ALL.
    InO
    has received attention as a bridging therapy before transplantation due to its high complete remission (CR) rate. However, the significance of
    InO
    in non-transplant patients remains unclear. We retrospectively evaluated four non-transplant patients treated with
    InO
    . All cases achieved CR after receiving at least two cycles of
    InO
    . Three of the four cases survived for more than 11 months without relapse. Moreover, all patients received
    InO
    as outpatients, because the adverse events were well-controlled.
    InO
    therefore appears to be a beneficial treatment even for non-transplant patients.

    The land-survey expedition conducted by
    Ino
    Tadataka over all parts of Japan during 1800-1817 was a monumental achievement in the Japanese history of scientific map making. Although almost all of
    Ino
    's diaries, field books and surveying instruments have long been preserved at the
    Ino
    Tadataka's Memorial Museum (Katori City), the sole missing is the meridian transit instrument for astronomical observations. Around 2003 a set of an old astronomical quadrant and a transit instrument was discovered, which was precisely produced and very similar, in terms of the structure, function and size, to the ones used by
    Ino
    at the Setonaikai district. The inscription recorded on the quadrant reads that it was produced in 1831 by Tatsunosuke at Karoto, a small harbor village of Kurahashi island near Kure city, with the help of an official astronomer of the Hiroshima han (fief), who was responsible for designing and inscribing the quadrant's scale. It is thus inferred that the two instruments were elaborate products which replicated
    Ino
    's ones by utilizing the chance when
    Ino
    visited Karoto in March of 1806. Hence the Tatsunosuke's quadrant and transit instrument are historically very valuable remains, the latter in particular, since it has been lost at the
    Ino
    's museum. The drawing "Yachu sokuryo no zu" preserved at the Irihuneyama Memorial Museum of Kure city has well been known by that this describes most vividly the practice of astronomical observations of the
    Ino
    's team with the relevant audience. Scrutinizing the historical local materials with drawings and explanations written in
    Ino
    ' diaries that recorded his expedition near the Kure region, we have found it most likely that a local painter drew "Yachu sokuryo no zu", who attended the
    Ino
    's night observations at Karoto. Motivated by this finding, we made twice extensive interview investigations at Karoto. As a result we succeeded in obtaining a testimony from the aged widow of an old traditional hotel whose owner's name in the 1810s was exactly the same as the one written in the
    Ino
    's diary. We also found out that the location of the hotel coincided with the one expressed with an asterisk, as a site of astronomical observations in the
    Ino
    ' large map of this region. Therefore, Tatsunosuke's astronomical instruments are important existence as central witness that combine the description of the
    Ino
    's diary, our findings at Karoto and the instruments themselves.

    背景)2015年に心臓外科周術期の肺高血圧症への一酸化窒素吸入療法(

    iNO
    )が保険償還されたが、本邦の心臓移植後におけるその有用性のデータは限られている。

    方法)当院で施行した178例の心臓移植のうち、当院で

    iNO
    が使用可能となった2016年以降の成人心臓移植95例が対象、術後の
    iNO
    使用の有無で比較した。

    結果)

    iNO
    vs 非
    iNO
    群で16 vs 79例。投与時間の中央値は20.4時間、年齢/性別/原疾患/移植前心機能/虚血時間に有意差なし。
    iNO
    群で移植前PDE-V阻害薬の内服が多く(44 vs 14%, p=0.015)、移植後1週の肺血管抵抗が高かった(1.6 vs 1.3単位, p=0.045)。
    iNO
    群で移植3日後のSOFAスコアが高い傾向あり(7 vs 6点, p=0.052)、挿管時間も長いが(20 vs 14時間, p=0.002)、ICU滞在日数や(7 vs 7日, p=0.311)、生存率に有意差なし。

    結語)移植後

    iNO
    使用率は約17%であり、肺血管抵抗の高い症例で使用される傾向あり。
    iNO
    使用の有無による生存率の差はない。

    Ino
    maps of the final edition completed in 1821 cover the whole Japanese Islands including Hokkaido and the belonging islands. However,
    Ino
    Tadataka surveyed the southern coast of the Hokkaido only once 1800. Comparing the final edition map and the map completed in 1804 which was drawn only by his own survey, contribution by
    Ino
    can thus be separated.

    We conclude that the results by

    Ino
    , for example, azimuth measurements to mountain summits and traverse line surveys along coasts, although these two are main parts of
    Ino
    works, are not used in the map of Hokkaido of the final edition. Place names too were newly collected by someone. Only the latitude values obtained by
    Ino
    with astronomical measurements of culmination altitude of stars were used for the final edition.

    In the yeast Saccharomyces cerevisiae, the expression of phospholipid biosynthetic genes, including the
    INO1
    gene (encoding inositol-l-phosphate synthase), is coordinately regulated by a cis-acting transcriptional element, UASINO (inositol-sensitive upstream activating sequence). For this paper we studied the effect of SCS2 disruption on
    INO1
    expression. SCS2 encodes a type II membrane protein and its deletion leads to inositol auxotrophy at temperatures above 34°C. We found that the expression of the
    INO1
    gene was reduced in the scs2Δ strain even when the cells were cultured under derepressing conditions for
    INO1
    expression. However, the β-galactosidase gene fused with the
    INO1
    promoter region was expressed normally in the scs2Δ strain. The phospholipid composition of scs2Δ cells was not dramatically changed compared with wild-type cells at 28°C, but the phosphatidylinositol level was reduced in scs2Δ cells cultured at 34°C. In addition, elevated phosphatidylcholine synthesis through the CDP-choline pathway was observed in the scs2Δ strain, and the disruption of genes involved in the CDP-choline pathway rescued the
    INO1
    expression defect of the scs2Δ strain. These results indicate that Scs2p can contribute to coordinated phospholipid metabolism including
    INO1
    expression by regulating phosphatidylcholine synthesis through the CDP-choline pathway.

    A) Tadataka
    INO
    (1745-1818) was a remarkable man who established a landmark in the history of cartography of Japan based on his indefatigable survey of the whole of Japan, carried out from 1800 to 1816. His work was so memorable and the Tokyo Geographical Society erected a bronze memorial monument in the Shiba Park, Tokyo, in 1889, but it was removed and lost during the World War II. However, the Society did succeed in the reestablishment of it in the same site with a beautifully designed stone monument in 1965.
    The account of his life and work was so remarkable that nearly twenty kinds of biographies and biographical stories of Tadataka
    Ino
    have been published since the Meiji era. Most of them placed the emphasis upon his indefatigable character, his laborious travel of survey throughout Japan (Fig. 4), and his successful achievement which means, the account of the life of Tadataka
    Ino
    has been didactically told in Japan, particularly in the elementary education before the war. Among them, however, “Tadataka
    Ino
    , the Japanese land-surveyor”, written by Prof. Ryokichi Otani (1917) is most substantial and authoritative, and some parts of which were revised or shortened, translated into English, and published in 1932.
    Prof. Otani scientifically analysed the
    Ino
    's survey and his manuscript maps and revealed their essential facts and the degree of accuracy of the maps. The book was reviewed by George Sarton in 1936 (Isis, XXXI, I, 196-200), and the account was briefly summarized in the article of Norman Pye and W. G. Beasley in 1951 (Geogr. Jour. CXVII, 178-187).
    Ino
    's remarkable achievement was appreciated in Europe earlier than in Japan. For instance, a map of Nippon compiled from
    Ino
    's map was published in Europe by von Siebold in 1840, with the result of demonstrating the Japanese progress in surveying and map-making. A somewhat similar kind of story which exemplified the accuracy of
    Ino
    's map was told in the article of Pye and Beasley, concerning manuscript copies of
    Ino
    's maps carefully kept in the National Maritime Museum at Greenwich. While in Japan, the Tokugawa Shogunate Government tucked the
    Ino
    's original manuscript maps (more than two hundred sheets of large-scale, middle-scale, small scale and others) in the Library, without using them effectively and closed to the public.
    B) Since the Meiji Restoration
    Ino
    's maps have served greatly to meet the urgent needs of modern maps, charts and statistical figures of the Japanese Islands. For instance, Military Land Survey Department produced hastily the maps of 1 : 200, 000 based on
    Ino
    's maps (Fig. 6), and some of the sheets served to the public up to 1920's before the modern maps replaced them out. Geographical Bureau of the Home Affairs provided statistical figures of the size of Japanese islands and prefectures and some of the figures maintained their existence up to 1921. Naval Hydrographic Department produced many charts based on the well drawn coastline of
    Ino
    's maps (Fig. 5), which have helped the development of coast navigation in Japan.
    In this way, Japan has owed the rapid progress in social and cultural development since the Meiji Restoration unconsciously and basically to
    Ino
    's maps ; in other words, without
    Ino
    's maps Japan would have met with various kinds of grave obstacles in the process of modernization. Many of the original manuscript of
    Ino
    's maps were lost by the fire in 1873 and the Kanto earthquake in 1923, but carefully copied maps, which provided the basis for practical use, are kept in the Library of the Geographical Survey Institute and others.
    The year 1967 falls on the 100th year since the Meiji Restoration and gives us a good opportunity to remember and to re-appreciate the remarkable achievement of, and the scientific geographical heritage from Tadataka
    Ino
    .

    A dominant mutation of Saccharomyces cerevisiae, CSE1, caused a decrease in the expression of the
    INO1
    gene product, inositol-1-phosphate synthase. The residual activity was completely repressed by the addition of choline to the medium. A mutant carrying this mutation could not grow in the presence of choline unless inositol was added to the medium. Here we report a suppressor gene of the CSE1 mutation, SCS1(suppressor of CSE1), which was cloned by complementation of CS-E1 with a wild-type multicopy yeast genomic library. The cloned SCS1 gene contained an open reading frame which encoded 304 amino acid residues with a calculated molecular mass of 34, 234 Da, and the sequence coincided with that of the
    INO2
    gene. An scs1/
    ino2
    null mutant constructed by gene replacement was viable, but auxotrophic for inositol and choline, and used for determination of the mRNA levels of various phospholipid-synthesizing enzymes. In agreement with the reported data for
    ino2
    mutants the disruptant showed decreased expression of the
    INO1
    and PSS genes, which are known to be regulated by inositol and choline. In addition, we newly found that the disruption of SCS1/
    INO2
    also caused a decrease in the expression of the CKI, PEM1, and PEM2 genes, which we previously showed to belong to the inositol-choline-regulated gene family. These results confirm and strengthen the conclusion that the SCS1/
    INO2
    gene is required for expression of inositol-choline-regulated genes in phospholipid synthesis.

    Phosphonylation of inosine (
    Ino
    ) has been achieved using inorganic diphosphonate (DP) in aqueous solution. The optimum condition for the phosphonylation of
    Ino
    with DP is
    Ino
    : DP = 1 : 10, pH 11 and 10 °C. 2´-Phosphonylinosine and 3´-phosphonylinosine were synthesized with the total yield of more than 95 %. The phosphonylated products of
    Ino
    were stable at 10 °C and pH 9. The reaction mechanism of
    Ino
    with DP was discussed.

    The survey of the almost entire islands of Japan carried out by Tadataka
    INO
    (1748-1818) was a remarkable and successful enterprise in the history of Japanese cartography, and many biographies and biographical stories of
    INO
    have been published. However, among them “Tadataka
    INO
    ” written in 1913 by Prof. Ryokichi OTANI (1875-1934) was the only one in respect to the scientific, authoritative and substantial point of view.
    Prof. OTANI, as a physicist and geodesist, naturally devoted nearly two-thirds of his pages to the description of
    INO
    's method, instruments, the adjustment of his surveys and his method of map-construction, the accuracy of maps, etc. On the contrary, the description about his travels of survey is treated rather briefly and subjectively and, in consequence, there remain some criticism in his description from the strict historical point of view. For instance, OTANI pointed out some instances of scandals which occurred between lords of clans and
    INO
    's party in the survey of Echigo and Kaga, while he emphasised the friendly help of the feudal lord of Kagoshima, contrary to the expectation of some difficulty and hindrance in such a powerful clan.
    The author analyzed these circumstances from many old documents and presented proper interpretation and also revealed that the
    INO
    's party changed its character and increased its importance after
    INO
    was appointed as an official of the Tokugawa Government in 1804.

    A dominant, single nuclear gene mutation, CSE1, caused inositol auxotrophy in yeast cells. The inositol requirement was marked when choline was present in the medium. Inositol-1-phosphate synthase, the regulatory enzyme of inositol synthesis, is repressed by inositol, or more profoundly by a combination of inositol and choline in the wild type. In CSE1, the level of inositol-1-phosphate synthase was low and was greatly repressed on the addition of choline alone. In accordance with this,
    INO1
    mRNA encoding the enzyme was low even under the derepressed conditions and was profoundly decreased by choline in CSE1. But in the wild type, the addition of choline alone had little effect. An
    INO
    1-lacZ
    fusion was constructed and the control of the
    INO1
    promoter in CSE1 was studied. lacZ expression was repressed not only by inositol, but also by choline in CSE1, whereas it was repressed by inositol, but only slightly by choline in the wild type. CSE1 was unlinked to the
    INO1
    structural gene. Thus CSE1 was thought to be a regulatory mutation. Furthermore, when the CDP-choline pathway was mutationally blocked, choline did not affect
    INO1
    expression, indicating that the metabolism of choline via the CDP-choline pathway is required for
    INO1
    repression.

    結核菌を含めた病原性抗酸菌に特異的な活性作用機序を持った新薬の開発が求められている.我々は2010年に膜リン脂質のホスファチジルイノシトールの生合成経路がヒトと抗酸菌とで異なる事を発見した.相違点の鍵となる酵素がホスファチジルイノシトールリン酸(PIP)合成酵素である.この酵素は放線菌門の一部の細菌のみに存在し,ヒトやヒトの常在細菌には存在しない.この酵素活性を阻害する化合物を見つけることができれば病原性抗酸菌に特異的な新薬の開発に繋がる.PIP合成酵素活性測定には,基質としてアイソトープでラベルされた1l-イノシトール1リン酸(1l-

    -1P)が必要である.この基質は市販されていないので,これまでは[14C]グルコース6リン酸([14C]Glc-6P)からメタン生成古細菌の粗酵素を1l-

    -1P合成酵素として使って調製していた.この粗酵素の活性は低く,粗酵素中の不純物の影響で生成物の1l-

    The effects of a novel piperazine derivative (
    INO
    2628) on sinus node pacemaker activity and atrial contractile force were investigated in isolated, blood-perfused dog atrium. Injections of
    INO
    2628 (0.03-100μmol) into the sinus node artery of the isolated atrium induced a dose-dependent decrease in sinus rate and an increase in contractile force. The positive inotropic effects at more than 10μmol were accompanied by a transient negative inotropism. Propranolol did not affect the positive inotropic response to
    INO
    2628, but it significantly suppressed positive chronotropic and inotropic responses to norepinephrine. Atropine at a dose which completely blocked negative chronotropic and inotropic responses to carbachol, produced a slight but significant depression of
    INO
    2628-induced negative chronotropic responses; inotropic responses were unaffected. These results suggest that
    INO
    2628 induces noncholinergic negative chronotropic and nonadrenergic positive inotropic responses in isolated dog atria.

    BACKGROUND

    Systematic reviews and other studies have not supported the routine use of inhaled nitric oxide (

    iNO
    ) for preterm infants with respiratory failure. However, despite the lack of evidence, many physicians use
    iNO
    for a subgroup of preterm infants with respiratory failure, such as cases of persistent pulmonary hypertension of the newborn (PPHN). This study aimed to clarify whether the use of
    iNO
    improved the mortality rate among preterm infants with PPHN.

    METHODS

    We analyzed data from the Neonatal Research Network in Japan, which is a population-based nationwide registry. The study included neonates who were born at <32 weeks of gestational age between 2006 and 2012 and were diagnosed with PPHN without major congenital malformation including serious congenital heart disease. The primary outcome was the in-hospital mortality rate. Confounder-adjusted odds ratios were estimated using mixed effects logistic regression accounting for clustering within hospitals as random effects and covariates as fixed effects.

    RESULTS

    We identified 1,231 eligible infants, including 739 infants (60%) who received

    iNO
    .
    iNO
    did not significantly improve mortality (odds ratio [OR]: 1.13, 95% confidence interval [CI]: 0.82–1.55, P = 0.45).
    iNO
    was significantly associated with bronchopulmonary dysplasia (OR: 1.50, 95% CI: 1.11–2.02, P = <0.01) and retinopathy of prematurity requiring treatment (OR: 1.56, 95% CI: 1.11–2.19, P = 0.01).

    CONCLUSIONS

    This retrospective study failed to detect an association between

    iNO
    treatment and improved survival among preterm infants with PPHN, however, there are several limitations in this study. Further research is required in this area.

    Background Congenital heart disease patients who have pulmonary hypertension (PH) require an evaluation for pulmonary vascular reactivity before surgical repair. In the present study the acute pulmonary vasodilating effects of 100% oxygen (O2), beraprost sodium (BPS) and 40 ppm inhaled nitric oxide (
    iNO
    ) during cardiac catheterization were compared. Methods and Results There were 90 patients who underwent cardiac catheterization for evaluation of PH (mean age, 16.5±16 years). The baseline mean pulmonary artery (mPA) pressure was 69.6±14.8 mmHg and the pulmonary arteriolar resistance (Rpa) was 13.8±8.3 Wood unit m2. Change in pulmonary vascular reactivity was defined as a decrease in mPA or Rpa >20% from baseline. The response to 100%O2,
    iNO
    and BPS during cardiac catheterization was 84%, 72.7% and 64%, respectively. Pair comparisons among each hemodynamic parameter showed no difference between the acute vasodilating effect of BPS and
    iNO
    . In some patients BPS showed a stronger effect than
    iNO
    in lowering Rpa. Conclusions BPS has a similar pulmonary vasodilating effect to
    iNO
    and can be used as an acute pulmonary vasodilating agent during cardiac catheterization with potential benefits over
    iNO

    Saccharomyces cerevisiae ire15 mutant has a defect in the expression of
    INO1
    , showing the inositol auxotrophic phenotype [Nikawa, J. (1994) Gene 149, 367-372]. We have isolated five yeast genes which suppress the ire15 mutation in multiple copies by genetic complementation. Among them, one gene, designated as SCS2, also suppressed the cholinesensitive dominant mutation, CSE1 [Hosaka, K. et al. (1992) J. Biochem. 111, 352-358]. The CSE1 mutation is not allelic to ire15. Sequencing analysis revealed that the SCS2 gene encodes 244 amino acid residues with a calculated molecular mass of 26, 866.

    /SCS1, which is another suppressor gene for CSE1 and is known to be a positive factor for
    INO1
    expression, also suppressed the growth defect of the ire15 mutant. These results clearly indicate that the ire15 and CSEI mutations genetically interact and the SCS2 and
    INO2
    /SCS1 genes are involved in the regulation of
    INO1
    expression.

    Genes involved in the phospholipid synthesis of Saccharomyces cerevisiae, such as PEM1, PEM2, PSS, and
    INO1
    , are coordinately repressed by myo-inositol and choline. In order to investigate this regulation, we transformed wild-type yeast with a PEM1 promoter-lacZ fusion and isolated two mutants, named ric1 and ric2 (_??_egulation by myo-_??_nositol and _??_holine), exhibiting decreased PEM1 expression. The lowered PEM1 expression in the mutants was monitored in colonies in terms of their failure fully to develop blue color on 5-bromo-4-chloro-3-indolyl-β-galactopyranoside-containing agar. ric1 mutant was auxotrophic for myo-inositol, indicating that
    INO1
    expression was also affected, whereas ric2 mutant required myo-inositol only in the presence of choline. The RIC1 gene was isolated by complementation of the
    Ino
    - phenotype of ric1 mutant and its identity was confirmed by genetic cross between the original ric1 mutant and a gene disruptant. The RIC1 gene was sequenced and found to be identical with the previously identified gene, SNF2/SWI2/GAM1/TYE3, which is known to encode a general transcription factor required for the expression of various genes including
    INO1
    . Analysis using various lacZ fusion constructs containing promoters for genes in phospholipid synthesis revealed that the expression of myo-inositol-choline-regulated genes, PEM1, PEM2, PSS, CKI, and
    INO1
    , was markedly decreased in the snf2/swi2/gam1/tye3/ric1 background, but the expression of a constitutive gene, PIS, was not. We conclude that SNF2/SWI2/GAM1/TYE3/RIC1 is a positive regulatory gene required for the expression of not only
    INO1
    gene, but also of myo-inositol-choline-regulated genes in general.

    The generational changes in the relative abundance of rice dwarf virus (RDV)-infected insects in a population of the green rice leafhopper, Nephotettix cincticeps were studied in relation to the amount of the virus-infected rice plants as sources for feeding aquisition. Investigations were conducted in two localities in Kochi Prefecture : Nangoku and
    Ino
    . Nangoku represented an area where early and late rice crops are cultivated, while
    Ino
    is an area of single crop of middle-season rice. Surveys were carried out during the period 1964-69 in Nangoku and during 1968-69 in
    Ino
    .In Nangoku, changes in the percentage of infected insects from generation to generation formed a parabolic curve with its peak at the 1st generation. In
    Ino
    , on the other hand, the percentage of infected insects decreased from the overwintered to the 2nd or the 3rd generation and again increased toward the 5th generation. The proportion of infected insects (P) had no significant correlation with that of infected rice hills. However, P was closely related to the index of infected rice plants (Iv) which was derived from the proportion of the biomass of infected rice plants to the total biomass of the rice plants in the field. In Nangoku, the relationship between P and Iv can be representedas follows : log P=0.727 log Iv+0.106, or P=1.278 Iv0.727.Whereas in
    Ino
    no relationship could be obtained because of insufficient data.

    The true north and magnetic north are not drawn in the maps of 1:36000 made by

    INO
    Tadataka at the beginning of 19th century. In this paper, the author developed method of estimation for the geographic meridian and magnetic meridian for digitized
    Ino
    maps. Correlation index between longitudes in the actual digital map of the Geospatial Information Authority of Japan and coordinates in the rotated map image becomes the maximum when the north of rotated map matches to the true north. In this way we were able to fix the north in
    Ino
    map of 1:36000.

    Ino
    had believed that the magnetic north was equivalent to true north anywhere through surveying and making maps of Japanese Islands. Therefore, north-south direction in his maps of 1:216000 is magnetic meridian which crosses parallels drawn at right angles. Notwithstanding lack of parallels in the map of 1:36000, we were able to fix the magnetic north comparing with the parallel of the map of 1:216000. This method is the same as true north fixing.
