再生医療のための培養軟骨細胞に対する機械的振動の影響

It was reported that cells show adaptive response by sensing mechanical vibration. The mechanism of it is not clear but this phenomenon can be applied to regenerative medicines of articular cartilages. In this study, we cultured chondrocytes from articular cartilages of Japanese white rabbits without any scaffolds and made cell sheets of chondrocytes. We prepared non-vibration groups and vibration groups for comparison and they were incubated for 20 days in order to make cell sheets of chondrocytes. The condition of mechanical vibration was sinusoidal wave of 25 Hz and 0.5 G. To observe the effect of cell proliferation and production of cartilage matrices, we measured the numbers of cultured chondrocytes and conducted an immunohistochemical examination with staining collagen type II and proteoglycan and compared between cell sheets of the non-vibration and vibration groups. As a result, the cell sheets of chondrocytes were made by cultivating 20 days in not only the non-vibration groups but also the vibration groups. However, the numbers of cells were not different between the non-vibration and vibration groups during 20 day cultivation. In the result of immunohistochemical examinations, there are no obvious differences in staining of collagen type II and proteoglycan between the non-vibration and vibration groups.
