In May 2010, severe symptoms caused by Soil-borne wheat mosaic virus(SBWMV)were observed in winter wheat fields(Triticum aestivum, cvs. Hokushin, Kitahonami, and Kitanokaori)in the central area of Hokkaido. The symptoms on leaves were observed as light green to yellow mosaic, and the growth of symptomatic plants were markedly stunted. SBWMV was detected from the diseased plants by tripleantibody sandwich enzyme-linked immunosorbent assay(TAS-ELISA)with monoclonal antibodies(Agdia Inc. and Neogen Inc.).The presence of SBWMV was further verified by reverse transcription(RT)- PCR using SBWMV universal primers SBWMV-UNIF and SBWMV-UNIR. The virus was transmitted from diseased plants to wheat seedlings(cv. Hokushin)through Chenopodium quinoa by mechanical sap inoculation, and detected again. This is the first report on the occurrence of SBWMV in winter wheat in Hokkaido.
