Cells of Salmonella typhimurium were killed through freeze-thawing in a ternary solution including NaCl, EtOH and water. The bactericidal effect of freezing was influenced by the solute ratio, NaCl/EtOH (R), and was maximum at R=ca.70/30. DSC-thermogram of the frozen solution in warming process showed a remarkable exothermic heat flow only in the solution whose R was 60/40-90/10. Extracellularly concentrated salts in the course of slow-freezing might injure bacteria in combination with ethanol which modified the permeability of membrane for water.

Thermal behaviors of water in the aqueous solutions of tetraalkylammonium bromide have been investigated through freezing and thawing processes by using low temperature DSC. Two different types of clathrate hydrates in the aqueous solution of tetrabutylammonium bromide were observed at 12℃ and 18℃ respectively by calorimetric method. In the solutions of tetramethyl, and tetraethylammonium bromide, formation of eutectic mixtures was observed at -13℃ and -22℃, respectively. They are independent of the concentration of solutes. Roles of molecular space in the aqueous solutions were discussed in relation to freezing of solutions.

Experiments were conducted to determine the transport properties of the dried layer using a radiant heating upon the sample surface. The samples of 6-30% coffee solution were freeze-dried in a dish of 70.5mm in diameter and 25 mm in height at constant surface temperatures ranging from 20 to 55℃ under about 10-95 Pa total pressure range, the usual operating condition of commercial freeze-dryer. Thermal conductivities and permeabilities of the dried layer were determined by applying the drying data to a model based on heat and mass transport in the sample. Thermal conductivities had a tendency to decrease as the sample concentration was increassed, and there was no definite effect of the dried layer temperature on thermal conductivity. Permeability decresced with increasing the sample concentration and it increased as the dried layer temperature increased. These transport properties were not affected by the pressure of water vapor in the dried layer under experimental conditions. From these results it may be concluded that transport properties depend mainly on concentration of sample tested and the concentration was one of the most important factors which affected the structure as well as transport properties of the dried layer. The relationships between transport properties and controlling factors were discussed in connection with the freezing and freeze-drying operations of the sample.

The infectivity of purified southern bean mosaic virus (SBMV) in 10mM sodium phosphate buffer, pH 7.0, decreased to 11% after freeze-drying. Surcose density gradient sedimentation profiles and electron microscopy clearly showed partial alteration of virions by freeze-drying. And such virions were found to be susceptible to RNase or sodium dodecyl sulfate. However, RNA isolated from such virions possessed the infectivity to the same level of RNA isolated from non-freeze-dried virions. Virions were protected by addition of various additives prior to freeze-drying. Upon the storage at 65℃, the preparation of freeze-dried virus lost the infectivity in one day. RNA isolated from such virions were degradated, and had no infectivity. Whereas virions with 0.5% lysine were retained the conformation, and mantained higher infectivity than the preparation with no additive. Also, the infectivity of RNA was gradually lost upon storage. Freeze-dried carnation mottle virus upon the storage at 65℃, showed the similar tendency to SBMV.
