Up to now, development of non-tray system freeze-dryers has been tried in many cases, but did not still reach the practical level for stable production, mainly due to the product loss in operation (or dried product remains inside the equipment in the form of dispersion or in adhesion) as pointed out by Lorentzen. Having restudied the methods by Mason and Seffinga, author has succeeded in developing a closed system for continuous freeze-drying of liquid materials by the means that an ice-lining is first placed on the inner (or outer) surface of a vertical tube and then frozen layer of liquid material is formed on the lining. Such ice lining is proved to be much effective not only in preventing dry product remains from being adhered to the tube surface, but also in eliminating the formation of isolated ice crystals and impermeable surface layer which impede drying.

We developed a new CRYO-UNIT which was directly connected to the scanning electron microscope equipped with a cold stage. This unit allowed to continuously observe the same frozen samples by both Cryo-SEM and freeze-replica which were prepared under exact temperature control with prevention of contamination. A few examples for the application of this unit to frozen biological samples were presented.

The freezing conditions of several plant tissue cultures, which have been preserved by subculturing of callus on solid medium were investigated in order to establish the method for long-term preservation. The cultured plant cells used were Datura innoxia, Rosa hybrida, Daucus carota, Nicotiana tabacum and Vinca rosea. They were cultured at 27℃ in dark for a week and used for the following studies. When the cooling rate was lowered than 1℃/min, the viability was not increased. Nor the hardening at 4℃ for 4 weeks was effective. The viability was high when cells were frozen in the solution of cryoprotectants, Murashige and Skoog medium + 5% DMSO + 10% glucose + 0.6M mannitol. When the viability and recovery growth were examined after storage at -80℃ and -135℃ in the mechanical freezer and at -190℃ in the vapor phase of liquid nitrogen, the storage at -135℃ seemed to be the best. The viabilities of the protoplasts of Datura innoxia and Vinca rosea, which were freeze-sensitive in the form of callus, were about 4 times and 17 times higher, respectively than that of callus after freezing to -40℃ at a cooling rate of 1℃/min and storaged at -190℃ for 18 hr.

The low-temperature thermal effect of glucose added to saline aqueous solutions was investigated by using differential scanning calorimetry(DSC). We obtained DSC data indicating that these mixed solutions can remain unfrozen during the cooling period and get crystallized by subsequent warming.
