The root fungal flora, soil microbial biomass, and soil microbial respiratory activity of 14 agricultural fields in which spinach was continuously cropped were investigated in the summer. Greenhouse cultivation has been carried out in this area, and the main nutritional component has been pig manure compost. The applications of pig manure compost were 30-120 t ha^<-1> year^<-1>. Poor spinach stands occurred in 25-80% of the cultivated fields. Neither the characteristics of soil chemistry nor the density of the root fungi showed a significant single correlation with the ratio of the area with a poor stand. However, multiple regression models including microbial respiratory activity and soil microbial biomass as predictor variables were obtained. To increase the microbial biomass, water soluble potassium and carbon and small amounts of nitrate would be necessary. In the field, where pig manure compost was applied excessively, soil chemical deterioration such as salt accumulation and imbalance of fertilizer component, caused by the storage of fertilizer component and accompanying anion component. Furthermore, there was some biological deterioration such as diminished biomass and respiratory activity. This outcome appears to have contributed to the pathopoiesis of soil-borne pathogens.

One hundred soil samples were collected from the Okinawa Islands, and soil properties relating to the nutrient supply capacity were evaluated. Among the 4 major soil types (Kunigami mahji, Shimajiri mahji, Jahgaru, and Daito mahji), Daito mahji showed significantly (p<0.05) higher labile organic carbon content than the others. However, Daito mahji showed the lowest value (15.3 ± 6.2 mg kg^<-1>) of available nitrogen. No significant relationships were observed in Daito mahji between the labile organic carbon and available nitrogen contents, in contrast to previous studies. The ratio of labile to total organic carbon of Daito mahji was significantly (p<0.05) higher than that of the other soil types, and a significant negative correlation was shown between the ratio and soil pH, suggesting that the lower pH in Daito mahji suppressed the decomposition of the labile organic matter and resulted in higher contents of the labile organic matter. An incubation study using Daito mahji showed a highly (p<0.001) significant positive correlation between the decomposition rate of added sunn hemp and soil pH, and liming significantly increased the decomposition rate (p<0.05). These results revealed that Daito mahji with higher contents of labile organic carbon could have potentially higher available nitrogen, although this would be impeded by soil acidity. Therefore, liming is a reasonable way to enhance soil productivity in sugarcane cultivation through the improvement of the ability of Daito mahji to decompose organic matter.

The application of organic matter, including compost, is known to improve the chemical, physical, and biological properties of soil. There are few studies on the biological improvements obtained from such application. By analyzing the PLFA and quinone, we investigated the changes in the soil microbial community structure in the field after continuous application of livestock waste compost. The total concentration of PLFA and the Shannon index H' of PLFA were high in the field after continuous application of livestock waste compost. The content rate of 18:2ω6,9 (mol%) was remarkably high in chemically fertilized fields. The total concentration of quinone and the Shannon index H' of the quinone profile were high in the field after continuous application of livestock waste compost. The content rate of Q7 and Q10 (mol%) was remarkably high in chemical fertilizer fields. The results of cluster analysis and principal component analysis based on the PLFA composition or quinone composition showed that the PLFA composition was greatly influenced by the application rate of organic matter and the quinone composition was greatly influenced by the type of organic matter applied.
