    Although abscisic acid-β-D-glucopyranosyl ester (ABA-GE) had been known to be a physiologically inactive conjugated ABA, the putative compound causing the allelopathic effect of Citrus
    junos
    fruit peel was isolated and the chemical structure of the inhibitor was determined as ABA-GE. In addition, an ABA-GE-cleaving enzyme, apoplastic β-D-glucosidase, which releases free ABA from the ABA-GE conjugate pool in the plant tissues, was recently found. In this research, growth inhibiting activity of C.
    junos
    , C. unshiu and C. hassaku fruit peel and ABA-GE concentration in their peel were determined, and a good correspondence between ABA-GE concentrations in C.
    junos
    , C. unshiu and C. hassaku fruit peel and the inhibitory activities of their aqueous methanol extracts was found. These results coupled with the effectiveness of ABA-GE on inhibition of growth suggested that ABA-GE may play an important role in the allelopathic activity of C.

    Abscisic acid-β-D-glucosyl ester (ABA-GE) was recently found in Citrus

    , C. unshiu, C. sudachi fruits and leaves. The concentrations in fruits of C.

    increased rapidly from the end of October, but the concentrations in the others did not increase so. The inhibitory activities of aqueous methanol extracts obtain from fruits and leaves of C.

    , C. unshiu, C. sudachi also correlated well with their ABA-GE concentrations.

    The allelopathic potential of an aqueous methanol extract of Citrus
    junos
    leaves was investigated under laboratory conditions. The extract inhibited the growth of roots and shoots of cress (Lepidium sativum L.), lettuce (Lactuca sativa L.) and alfalfa (Medicago sativa L.) seedlings. Significant reductions in the root and shoot growth were observed as the extract concentration increased. Since abscisic acid -β-D-glucopyranosyl ester (ABA-GE) was found to be a major inhibitor in fruit peel of C.

    , ABA-GE in C.
    junos
    leaves was determined and the concentration was 540 μg kg-1 fresh weight in the leaves harvested on September 26, 2002. These results suggest that C.
    junos
    leaves possess allelopathic activity and could work as a weed inhibiting agent, which should be investigated further in the field for practical application.

      Citrus juice processing residues are mainly composed of peel, juice sack and seed. The peel, especially, consists of bioactive compounds such as flavones. Supercritical carbon dioxide (SC-CO2) extraction of flavonoid was carried out at a pressure of 30 MPa and temperature ranging from 80 to 160°C. Ratio of water and ethanol as a co-solvent was varied from 0 to 100%. Flavonoids such as naringin and hesperidin have glycoside group, thus water was a good solvent for extraction of these compounds. Extraction behavior of tangeretin was quite different from those of flavonoid glycosides, and the yield of tangeretin tended to increase with ethanol concentration.

    養殖ブリの高付加価値化を目的として下記の試験を行った。試験 1 ではユズ果汁をブリ飼料に添加することによって血合筋の褐変を抑制できるか検討した。飼料 1 kg にユズ果汁を段階的に添加し,ブリ幼魚に 40 日間給与した。ユズ果汁の添加によって,成長を損なう事無く,血合筋の褐変が抑制されていた。試験 2 としてユズ果汁を添加した飼料を 30 日間与えたブリの筋肉中からユズ香気成分の検出と同定を行い,香りの成分が果汁を添加した飼料より移行し,蓄積されたことを明らかにした。

    ユズ,ナオシチ,ムカクユ,オオユ,ベニユ,ユコウ,スダチ,ダイダイ,スミカン,キヨオカダイダイ,トコス,ウジュキツ,タチバナの13種類の酸用カンキツ及びウンシュウについて有機酸,糖,ペクチン,アスコルビン酸の分析を行った。
    (1) 搾汁率はトコス,ムカクユ,ユコウで30%以上あり,一方,ユズ,スミカン,ウジュキツでは20%前後と低かった。
    (2) 有機酸分析の結果,9~15個のピークが検出された。主要な酸であるクエン酸とリンゴ酸を始め,乳酸,酢酸,ギ酸,ピルビン酸,フマール酸,t-アコニット酸の存在が認められた。クエン酸とリンゴ酸を合せた濃度はほとんど酸用カンキツで4~6%に達した。
    (3) 糖酸比の逆数は酸用カンキツの特徴を明瞭にし,ウンシュウ1.0に対して,ムカクユ,ユズ,スダチ,オオユが20以上,キヨオカダイダイ,ダイダイ,ユコウ,ナオシチ,トコス,スミカンが10~20であった。
    (4) アスコルビン酸含量は,酸用カンキツでは40mg%以上のものが多く,とくにオオユ,キヨオカダイダイ,トコスで高かった。

    A total of 39 aroma compounds were detected in the essential oil of Citrus nagato-yuzukichi Tanaka (nagato-yuzukichi) by gas chromatography-mass spectrometry (GC-MS). The essential oil was characterized by a high percentage of monoterpene hydrocarbons (12 components, 90.52%). The composition pattern of essential oil in C. nagato-yuzukichi was fairly similar to that of Citrus sudachi Hort. ex Shirai (Sudachi). Principal component analysis (PCA) of data obtained with an electronic nose indicated a variation of each oil along PC1. The oils of Citrus
    junos
    Tanaka (Yuzu) and Citrus sphaerocarpa Tanaka (Kabosu) showed a clear upward displacement as compared with those of C. nagato-yuzukichi and C. sudachi. However, in PC2, the oils of C. nagato-yuzukichi and C. sudachi showed a displacement in a negative direction and a positive one respectively.

    The isotope ratios of monoterpene hydrocarbons in Citrus
    junos
    Tanaka (yuzu) essential oils from different origins were determined by ordinary high-resolution gas chromatography-mass spectrometry (HRGC-MS). Both intensities of the molecular mass peaks (m/z 136) and of the isotope peaks (m/z 137) of monoterpene hydrocarbons were measured by single-ion monitoring with an MS analysis. The isotope ratios (m/z 137/136) of the ten monoterpene hydrocarbons commonly contained in citrus essential oils, α-pinene, β-pinene, sabinene, myrcene, α-phellandrene, α-terpinene, limonene, γ-terpinene, β-phellandrene and terpinolene, were determined in yuzu samples of the highest commercial quality from 42 different production districts. Statistical treatment of these data by the t-test and sign test revealed significant differences of the isotope effects in each yuzu sample. It is suggested that this technique will be applicable for evaluating the quality, genuineness and origin of citrus fruits and their products. The isotope fingerprints were also demonstrated in several citrus fruits other than the yuzu samples.

    Electrofusion was conducted to use the embryogenic callus cells of satsuma mandarin (Citrus unshiu cv. ‘Juman’ unshiu) and mesophyll cells of yuzu (C.
    junos
    ) and lemon (C.limon cv. ‘Eureka’ lemon). Two plants were regenerated from the combination of satsuma mandarin and yuzu, and one plant from that of satsuma mandarin and lemon. The plants had 18 chromosomes (2n=2×=18 in each parent), and showed the same nuclear rDNA fragment pattern as that of the mesophyll parent. The mitochondrial DNA analysis showed the presence of specific yuzu or lemon bands in addition to specific satsuma mandarin bands. From these results with morphological evidence, the regenerated plants were considered as cybrids with recombined mitochondrial DNA.

    Ulcerative colitis is a well-known inflammatory bowel disease. Although there are drugs that are effective against this disease, the prevention and attenuation of ulcerative colitis by food rich in functional ingredients without side effects is desired because some drugs have side effects. In this study, we investigated the effects of yuzu (Citrus

    junos
    Tanaka), a citrus fruit native to northeast Asia, on a mouse dextran sulfate sodium (DSS)-induced colitis model. Mice given drinking water containing DSS showed significant weight loss, colon shortening, diarrhea, and visible fecal blood. In contrast, mice fed a diet containing 5% yuzu peel for 14 d before receiving DSS showed significant attenuation of these phenotypes. To clarify the mechanism underlying the attenuation, we investigated the anti-inflammatory and antioxidant effects of yuzu peel. We found that yuzu peel extract suppressed tumor necrosis factor-α (TNF-α) production in lipopolysaccharide (LPS)-stimulated mice and murine macrophage cell line through suppression of nuclear factor-κB (NF-κB) activation. In addition, we confirmed that yuzu peel extract had a moderate antioxidant effect. These results suggest that yuzu peel attenuates the pathologies of DSS-induced colitis by coordinately suppressing inflammation and oxidative stress against lipids in vivo.

    Changes in 44 compounds of yuzu (Citrus
    junos
    Sieb. ex Tanaka) steam-distilled peel oil (SDO) and possible artifacts that accrue during storage at 25°C were investigated after 1, 2, 3, 4, 8 and 12 weeks. Changes in SDO composition during storage took place more rapidly than in cold-pressed oil. Total monoterpene hydrocarbons decreased markedly, with major losses of limonene and γ-terpinene and notable increases in p-cymene, as well as alcohols. Bicyclogermacrene, the main sesquiterpene hydrocarbon of the fresh oil, practically disappeared. (–)-Spathulenol, the main artifact, was converted from bicyclogermacrene. Antioxidants such as α-tocopherol, β-carotene and 2,6-di-tert-butyl-4-hydroxytoluene (BHT) slowed the formation of oxidation products, such as p-cymene, (–)-spathulenol by inhibiting the formation of oxidative products. This suggests that the antioxidants can be used to maintain yuzu oil quality.

    We cloned the gene of the acyclic sesquiterpene synthase, (E)-β-farnesene synthase (CJFS) from Yuzu (Citrus
    junos
    , Rutaceae). The function of CJFS was elucidated by the preparation of recombinant protein and subsequent enzyme assay. CJFS consisted of 1867 nucleotides including 1680 bp of coding sequence encoding a protein of 560 amino acids with a molecular weight of 62 kDa. The deduced amino acid sequence possessed characteristic amino acid residues, such as the DDxxD motif, which are highly conserved among terpene synthases. This is the first report of the cloning of a terpene synthase from a Rutaceous plant. A possible reaction mechanism for terpene biosynthesis is also discussed on the basis of sequence comparison of CJFS with known sesquiterpene synthase genes.

    The volatile flavor components of ripe and overripe ki-mikan (Citrus flaviculpus Hort. ex Tanaka) peel oil samples, which had been isolated by cold-pressing, were investigated by capillary GC and GC-MS, and compared with the Hyuganatsu (Citrus tamurana Hort. ex Tanaka) flavor. Limonene (ripe fruit, 82.44%; overripe fruit, 73.10%) was the most abundant compound in the ki-mikan oil, this being followed by γ-terpinene (8.83% and 13.74%), trans-β-farnesene (1.76% and 3.12%) and myrcene (1.54% and 1.13%). The composition of overripe ki-mikan oil was characterized by higher amounts of aliphatic and sesquiterpene hydrocarbons, monoterpene and sesquiterpene alcohols, ketones and esters than that of ripe ki-mikan oil. Monoterpene hydrocarbons, especially limonene (84.78%), were predominant in Hyuganatsu oil. The CPO composition of ki-mikan was qualitatively similar to that of Hyuganatsu, but differed quantitatively. The content of sequiterpene hydrocarbons was higher in the ki-mikan oil samples than in Hyuganatsu oil, while ketones showed the opposite predominance. These differences were more evident in the trans-β-farnesene and l-carvone contents. The ratio of both these compounds could be used to distinguish ki-mikan oil from Hyganatsu oil.

    The occurrence and distribution of flavanone glycosides in flowers, young and mature leaves, young and ripe fruits, dividing them into peels, segment membranes and juices, of many kinds of citrus were investigated. The occurrence of narirutin, eriocitrin and neoeriocitrin is fairly common to a number of species in genus citrus. Results also indicate that flavonoid patterns in citrus can be classified into eleven groups or more and further they can be summarized in two large groups: (a) those composed of flavanone rutinosides and (b) those composed of flavanone neohesperidosides. Thus the chemotaxonomic table of citrus was given as the results of our study in connection with the Tanaka's classification.
    Flavonoids in some artificial citrus hydrids were also studied.

    1. 1954年4月から8月に2回に亘つて(36日間と45日間),地温を20~22°C,30~32°C,40~42°Cとして,8種のカンキツ類実生の夏季の生育を比較すると,ナツダイダイ,ヤマミカン,法元ブンタンなどは地温30~32°Cにおいて,ユズ,カラタチ,タチバナ,日向ナツミカンなどは地温20~22°Cにおいて生育が最もよく,コウジは地温22°Cと32°Cとで生育が殆んど同じであつた。20~22°Cにおいても,ナジダイダイ,ヤマミカン,法元ブンタンの生長量は他に較べて多い。40~42°Cでは8種ともに生育が著しく劣り,ナツダイダイ,ヤマミカン,法元ブンタンのみ僅少ながら生長した。
    高知の夏季の地温は,地表下30cm以内で24~31°Cである。従つて,地温の点でナツダイダイ,ヤマミカン,法元ブンタンなどの実生が台木として適し,ユズがこれに次ぐ。カラタチは不適当で,台木として使用するならば深根になるように管理し,敷藁,覆土も必要である。
    2. 1955年2月7日から3月6日まで(水耕法)と1957年12月23日から翌年4月30日まで(普通のポット栽培)と2回に分け,地下部の温度を高めて,14種のカンキツ類実生の冬季から春季に亘る生育を比較した所,加温によつて,根の伸長開始期が著しく早くなり,1日当り伸長量も多く,伸長期間が延長された。地温18~23°Cで1月中旬に,地温9~15°Cで1月下旬~2月上旬に根が伸長し始め,地温3~13°Cに較べて,前者で45~56日,後者で26~38日も早い。概して地温23°C以下では,耐寒性のあるカラタチ,ユズ,コウジの根が早く,ハツサク,ダイダイ,舟床ミカン,日向ナツミカン,タチバナなどの根が前者らに次いで早く,耐寒性の余りない福原オレンジ,ワシントン・ネーブルオレンジ,ヤマミカンなどの根はおそく伸長し始める。地温上昇に伴なう根の伸長の促進程度は耐寒性に劣るものほどやや高い。
    3. 冬季における地下都の温度が8~13°Cであると,カラタチを筆頭にコウジ,ユズ,ダイダイ,舟床ミカンなどは特に生長が旺盛で,ナツダイダイ,日向ナツミカン,サンボウカンなども前者に次いで比較的生育がよい。ハツサク,ヤマミカン,法元ブンタン,タチバナ,福原オレンジ,ワシントン・ネーブルオレンジなどはいずれも生育が劣る。しかし,地下部の温度が3~13°C区に較べて,カラタチ,コウジ,ユズなどが2倍弱,ダイダイ,舟床ミカン,日向ナツミカン,サンボウカンなどが2~3倍,ナツダイダイ,ヤマミカン,タチバナなどが3倍強,オレンジ類がそれ以上で,耐寒性の余りないもめほど倍率が高い。高知の地表下50cm以上の地,温は冬季と雖も8.5°C以下に下らないが,本邦のカンキツの根は地表下30cm以上に殆んど及んでいない。従つて冬季の地温の点から,耐寒性のあるカラタチ,ユズ,コウジなどや深根性であるユズ,ナツダイダイなどが台木として適し,ダイダイ,舟床ミカンなども比較的よい台木といえる。なおカンキツ類の台木の根が深くなるように努め,地温保持の管理方法が一層望まれる。

    The characteristic aroma compositions of Citrus
    junos
    Sieb. ex Tanaka (yuzu) cold-pressed peel oil were studied by both instrumental and organoleptic methods. The samples were obtained from 5 popular yuzu production districts: Kochi, Tokushima, Ehime, Oita and Wakayama prefectures of Japan, at common harvesting periods early, middle and late November. Of 174 compounds detected, 67 compounds were identified and quantified. The compositional and olfactometry profiles of yuzu essential oils from Kochi were compared with those from the 4 other origins. The ratio of monoterpene to sesquiterpene hydrocarbons also presented the characteristic citrus aroma profile. Principal component analysis based on frequency of descriptive notes was carried out. Some difference was identified between the samples from Kochi and other origins, even those harvested in the most common harvesting period. In general, the profile of yuzu from Oita and Wakayama was approximately the same as that from Kochi.
