In this paper a simple method for chromosome preparations from amphibian intestinal epithelial cells is shown, making use ofthe Japanese frogs and salamanders. The procedures presented here in detail are as follows, 1) 2 mg/ml colchicine treatmentfor 3~24 hours, 2) 0.075 M KCl hypotonictreatmentfor 15~40 minutes, 3) fixation by a mixure of acetic acid and methanol (1 : 3), and 4) preparation of specimens by air-dried or squashed method. After chromosome staining, karyotypes of frogs and salamanders were analyzed by using the terminology for centromericposition as quantitatively.
