In order to discern
sake yeast strains, specific polymerase chain reactions (PCR) which targeted 22 ORFs containing the minisatellite sequences were tested. We found the difference in DNA fragments to be amplified by using the five pair primers designed
YOR009W (TIR4),
YDR150W (NUM1), YKR102W (FLO10), YDL037C (BSC1), and
YNL190W of sake yeast strains. Since
shochu yeast strains differed in PCR products by using primers for which
sake yeast strains showed no difference in the length of PCR products, the lower degrees of the variety of the amplified DNA fragments of
sake yeast strains were speculated to cause the genetic difference in these strains to be lower in comparison with other brewing yeast strains.
