The mycotoxin ochratoxin A (OTA) has antibiotic properties. By analysing these pro-perties, a competitive inhibition of phenylalanyl-tRNA synthetase (PheRS) was found. This type of enzyme was inhibited in the same manner in higher cells too. Increased concen-trations of phenylalanine, therefore, in vivo prevented several effects of OTA in animal systems. The reason of an inducible resistance to OTA of B. subtilis was found to be due to increased of PheRS. A ochratoxin A resistant constitutive mutant had an altered PheRS. Ochratoxin A and 4R-OH-ochratoxin A acted as immunosuppressors, whereas ochratoxin a did not.

A sensitive analytical method for two mycotoxins, luteoskyrin and rubroskyrin, was developed by using reverse phase HPLC. Luteoskyrin and rubroskyrin were eluted from ODS column with CH³CN :H²O (40:60) containing 0.5% CH³COOH. Detection was efficiently made by visible absorption at 429 nm. Neither ultraviolet detection at 254 nm nor fluorescence detection gave better result. Detection limit was about 10 ppb when the method was applied to rice sample.

Anthraquinone mycotoxins, averufin and versicolorins A, B, from Aspergillus versicolor (Vuillemin) Tiraboschi were found to be potent uncouplers on oxidative phosphorylation of isolated rat liver mitochondria, suggesting their strong cytotoxicities. It was revealed that these anthraquinone mycotoxins were the group of the strongest uncouplers among mycoto-xbs.s so far tested.

Duclauxin and related compounds were examined for cytotoxicities on murine leukemia L1210 culture cells and inhibitory effects on mitochondrial respiration. The potencies in cytotoxicity and inhibitory effect of duclauxin were markedly decreased when the oxygen atom of isocoumarin nucleus was substituted by nitrogen but were not significantly decreased when the oxygen atom was substituted by tertiary amines to produce N-methyl and Nethyl-duclauxamides, showing a good correlation between cytotoxicity on L1210 culture cell and inhibitory effect on mitochondrial respiration.

The effect of flavoglaucin, a benzoquinol pigment from Aspergillus chevalieri (Margin) Thom et Church, on mitochondrial respiration was studied using isolated rat liver mito-chondria. Flavoglaucin exhibited an uncoupling effect, causing marked decreases in RC index and P/O ratio of mitochondrial respiration. The ID₅₀ was around 20μM. Flavoglaucin induced drastic swelling of mitochondria at the uncoupling concentrations even in isotonic sucrose solution in which pseudo-energized swelling is inhibited. It was suggested, thus, that the induction of mitochondrial swelling might be responsible for the uncoupling activity of flavoglaucin, since flavoglaucin do not possess any dissociable group at physiological pH range.

Female mice were given a single injection of 5 to 30 μg zearalenone on the various days after birth. A high incidence (about 60%) of persistent vaginal cornification depending on the dose or the day of treatment was induced. Furthermore, all of the mice daily injected with 30 μg zearalenone for the first 5 days after birth showed persistent estrus which were not abolished by later ovariectomy, adrenalectomy or combined ovariectomy with adrenaletomy. Significant reduction of the mating rate and infertility were also observed in this group. These females were anovulatory showing no corpora lutea but a few large follicles in the ovaries. Neonatal zearalenone treatment led to a significant decrease in the number of the gonadotropes in the hypophysis.From the above-mentioned results, it seems likely that hypothalamus-pituitary system is affected by neonatal treatment with zearalenone.
