The effect of flavoglaucin, a benzoquinol pigment from Aspergillus chevalieri (Margin) Thom et Church, on mitochondrial respiration was studied using isolated rat liver mito-chondria. Flavoglaucin exhibited an uncoupling effect, causing marked decreases in RC index and P/O ratio of mitochondrial respiration. The ID
50 was around 20μM. Flavoglaucin induced drastic swelling of mitochondria at the uncoupling concentrations even in isotonic sucrose solution in which pseudo-energized swelling is inhibited. It was suggested, thus, that the induction of mitochondrial swelling might be responsible for the uncoupling activity of flavoglaucin, since flavoglaucin do not possess any dissociable group at physiological pH range.
