    Fundamental and clinical studies on AC-1370, a new antibiotic, were carried out in the field of ophthalmology and the following results were obtained.
    1. The MIC of AC-1370 against Staphylococcus aureus isolated from infectious eye diseases ranged from 12.5-100μg/ml, and MIC of AC-1370 against Klebsiella pneumoniae ranged from 1.56-50μg/ml.
    2. Aqueous humor concentration of AC-1370 reached maximum (2.6μg/ml) at 1 hour after an i.v. administration of 50mg/kg in rabbits, and the maximum serum concentration (21.0μg/ml) was obtained at 30 minutes after the injection.
    3. High concentration in tear and low concentration in aqueous humor were measured after i.v. administration of 50mg/kg AC-1370 in rabbits.
    4. Aqueous humor concentration of AC-1370 in rabbits reached peak at 11 hour after subconjunctival injection 10mg/0.1ml and the peak aqueous humor level was 10.1μg/ml.
    5. Two patients with ocular suppurative diseases were successfully treated at a daily dose of 1, 000, 2, 000mg AC-1370. 6, No side effect were observed.

    In order to study oral surgical application of AC-1370, fundamental and clinical investigations were carried out and the following results were obtained.
    1) Rabbits were intravenously injected with AC-1370 at a dose of 33 mg/kg over a period of 2 hours. The levels of serum and submandibular gland attained to maximum at the end of injection, showing 76.35±6.017 pern1 and 6.69±0.607 peg respectively. The levels of tongue, gingiva, parotid gland, and cervical lymphonodi attained to maximum at 2 hours and 15 minutes from the begining of injection, showing 16.24±0.668μg/g, 30.23±4.770μg/g, 13.52±1.476μg/g and 17.26±0.406μg/g respectively. The ratios of such peak levels vs. peak serum level were in the following order: gingiva< cervical lymphonodi< tongue< parotid gland< submandibular gland.
    2) AC-1370 was intravenously administrated to 15 cases with infection in oral region at daily dose of 1 to 2 g for 1 to 7 days. The clinical effectiveness rate was 93%.
    3) Side effects were observed in 2 cases, which consisted of headache in one case, and rash and nausea with decrease of blood pressure in one case. No abnormal laboratory findings were noted.

    Clinical and laboratory studies of AC-1370, a new synthetic cephalosporin, were carried out and the following results were obtained.
    AC-1370 was administered to 10 patients of acute infections of oral and maxillofacial regions consisting of 7 cases of mandibular ostitis, 2 cases of cellulitis of floor of mouth and 1 case of cellulitis of cheek, at a daily dose of 2 g for 3-7 days. The results based on the points determined byithe criteria recommended by the Japanese Society of Oral Surgery revealed excellent in 2 cases, good in 6 cases, poor in 2 cases. No side effect was observed.
    The sensitivity was determined by the plate dilution method with 16 strains of anaerobic Streptococcus and 23 strains of aerobic Streptococcus that were isolated from acute infections of oral and maxillofacial resions. The growth of 50% of Streptococcus was inhibited at concentration of less than 6.25μg/ml and all of the strains was inhibited at concentration of less than 25μg/ml.
