    The authors devised a technique to reach the chiasm via the transsphe-noidal route in mongrel dogs. When this technique is employed, a chiasmal compression model by a small balloon can be obtained without performing craniotomy.

    Reactivity of natural killer (NK) cells to an established human renal cancer cell line (KU-2) was determined by cellular immunoadsorbant tech-nique, utilizing human leukemia cell line (CCRF-CEM) which is highly sensi-tive to NK cells. The influence of human alpha interferon (HuIFN-α) on NK activity against KU-2 cells was also determined by microcytotoxicity assay. Percent cytotoxicity increased 84%, 52% and 59% in non E-rosette forming (non ERF) lymphocytes, ERF lymphocytes and unseparated peri-pheral blood lymphocytes (PBL), respectively, under effector cells/target cells ratio being 25/1 and preincubation with 150 IU/ml HuIFN-α. The re-sults indicated that more marked influence of HuIFN-α upon non ERF lym-phocytes was expected than ERF lymphocytes.
    Identification of the effector lymphocytes under influence of HuIFN-α was undertaken by scanning electron microscopic observation. Lymphocytes characterized by relatively smooth surface and short microvilli were observed to be adherent to the surface of the KU-2 cells showing destruction of their morphological integrity.
    To determine the direct cytotoxic effects, the KU-2 cells were exposed to HuIFN-α. When exposed to 50IU/ml HuIFN-α, cellular doubling time extended from 23 hrs to 38 hrs. With more than 200 IU/ml, the growth was acceptably inhibited and no dose dependent effect on the growth curves was observed in large dose series.
    From the results of in vitro studies, it was determined that both NK activation and direct inhibitory mechanisms were involved in the growth inhibitory effect of HuIFN-α against KU-2 cells. HuIFN-α was suggested to play an important role in a multimodal treatment of human renal cell carcinoma.

    To perform efficient arteriography for patients with arterial occlusive dis-ease of the lower limbs, a new system for serial whole-limb arteriography with extra-large-film changer was developed and clinically applied. This system consists of a motor driven cassette-film changer with four pieces of film sized 35.4×106.2 cm per frame, and ancillary fluoroscopic facilities. The source-image distance is 1.6m high to cover a wider field and to minimize radio-graphical blur. The film is 35.4 ×106.2cm, sufficient to cover the pelvis and both legs in average sized Japanese patients. The serial angiographic mode can be timed at 1 frame per 1.5 sec exposures at a shorter range and at 1 frame per 10 sec exposures at a longer range. A intensifying screen and a equalized alminium wedge filter are adopted to obtain an adequate density as uniform as possible in the abdomen down to the calf. Last 2 years, the author performed 67 occasions of Aorto-arteriography for 46 patients with arterial occlusive disease of the lower limbs. They were 25 patients with arterioscle-rosis obliterans, 14 patients with thromboangitis obliterans, and 7 patients with other arterial occlusive disease. Excellent results were obtained with our new system in serial whole-limb arteriography.

  • ON-OFF MECHANISM BY LH-RH

    To clarify the on-off mechanism of gonadotropin synthesis and its release in the anterior pituitary gland, the anterior pituitaries of the male rat were cultured in vitro through a leuteinizing hormone-releasing hormone (LH-RH) stimulation and a washing sequence for the analysis of cyclic nucleotide and gonadotropins. After the first stimulation of gonadotrophs with synthetic LH-RH for 20 minutes and the removal of LH-RH by washing, the alteration of the gonadotropin release and its content in the anterior pituitaries were investigated for several hours. The data indicate that the primary stimu-lation by LH-RH to the gonadotrophs has no continuous effect on the gonado-tropin release and synthesis after washing and that LH-RH, primarily ad-ministered, does not change the responsiveness to the secondarily administered LH-RH. The experiments also showed that by the stimulation of LH-RH, adenosine cyclic 3', 5'-monophosphate (c-AMP) was accumulated in the an-terior pituitary cells, but that such high accumulation did not continue after washing, and gonadotropins were released and synthesized by the admin-istration of dibutyryl c-AMP and theophillin. It is suggested that the con-tinuous gonadotropin release and synthesis required the continuous LH-RH stimulation, which maintains an elevated c-AMP concentration in the rat anterior pituitary gland.

    The author measured intrapleural pressure and pulmonary arterial wedge pressure simultaneously during quiet breathing and coughing in 10 post-thoracotomy patients by using a balloon-tipped catheter in the pleural space of the thoracotomy side and a Swan-Ganz flow-directed catheter in the ipsilateral or contralateral pulmonary artery. It was found that the pul-monary arterial wedge pressure during quiet breathing and coughing closely reflects the intrapleural pressure, and that the peak value of the pulmonary arterial wedge pressure at coughing reduced by the mean wedge pressure value during quiet breathing represents the intrapleural peak pressure. Therefore, it was considered that the pulmonary arterial wedge pressure tracing might be an acceptable alternative to the intrapleural balloon method or intra-esophageal balloon method for measuring intrapleural cough pressure. In the post-thoracotomy patients, the markedly decreased amplitude and peak value of the intrapleural pressure of the thoracotomy side compared to those of the non-thoracotomy side would seem to demonstrate the inefficiency of sputum expectoration from the tracheobronchial tree of the thoracotomy side.
