Sixteen mongrel dogs weighing 10-17kg were devided into three experimental groups, DHP, control and intraarterial infusion group. Hemoperfusion over charcoal was performed for 180 min with blood flow of 100ml/min immediately after administration of anticancer agent using Hydron-coated activated charcoal, DHP-1 (Kuraray, K. K., Japan). A large dose of anticancer agent, mitomycin C 1mg/kg, was administered into femoral vein in a one shot manner (infusion time was less than two minutes). In control group, the same dose of mitomycin C was administered into the antecubital vein without hemoperfusion. All animals died within 8 days after bolus administration of mitomycin C (1mg/kg) in control group due to acute intoxication of mitomycin C, on the contrary, all dogs were completely rescued from this toxicity by direct hemoperfusion. Distribution volume in the central compartment (V
1) was 0.17833±0.03587l/kg in control group and 0.45251±0.06843l/kg in DHP group (p<0.01). Distribution volume in the peripheral compartment (V
dss) was larger in DHP group than in control group, also (p<0.001). Estimated mitomycin C removed during hemoperf usion was 13% of the administered doses.

Scleritis were induced in about 170 patients, whose hemodialysis were performed with NAC-dialyzer composed of cellulose acetate hollow fibers. Though pain and redness of eyes persisted for about 2 weeks, visual disturbances improved completely except one case. Searches for etiologic agent were carried out. Extracts of the dialyzer injected intravenously to animals caused conjunctivitis in rabbits, and scleritis in dogs. Fractionations of extracts were performed by Sephadex LH-20. When each fraction was injected to animals, Fr. 5 and Fr. 6 produced redness of eyes. Analyses of the Fr. 5 and Fr. 6 by IR. UV and NMR demonstrated that these fractions contained cellulose acetate oligomers and urethane oligomers. Substances were produced experimentally by the reaction of cellulose acetate oligomer and urethane. Extracts of these substances showed in Sephadex fractionation just same Fr. 5 and Fr. 6, and injections of these fractions in dogs produced scleritis. These results indicate that causal factors are composed of cellulose acetate oligomer and urethane oligomer, and produced by reaction of the two at the assembly of dialyzer.
