    Localization and intensity of three lysosomal enzymes such as acid phosphatase, β-glucuronidase and N-aceytl-β-glucosaminidase in HeLa cells were examined during the course of cultivation. Acid phosphatase showed the most intense reaction. Localization and intensity of acid phosphatase were varied in different cell groups. Such varieties were classified as Type I to Type IV. Type I indicated young cells wherein stained granules were localized in the Golgi area. Type II indicated young cells with diffusely scattered, stained granules. Type III indicated rounded dying cells, and the whole cell was stained. Type IV indicated cells wherein the enzyme activity was almost negative. The quantitative activity of acid phosphatase was found to be high when young cells, Type I, grew in the earlier stage and when young cells, Type II, increased in the intermediate stage, and became lower as cells, Type IV, increased in the later stage. Thus, the lysosomal enzyme activity in HeLa cells seemed to consist of two factors; one is the primary lysosomes indicating cellular growth or cellular activity, the other is the secondary lysosomes indicating cellular degradation.

    Effects of glucocorticoids (triamcinolone diacetate (TAG) and dexamethasone) on the hepatocellular fructose-1, 6-diphosphatase (FDPase) activity was investigated histochemically in some rodents.
    It was observed that glucocorticoids induced the hepatocellular FDPase and this induction was heterogenous in nature, indicating the functional heterogeneity among hepatic cells. This phenomenon was named as “TAC effect”. The TAG effect is not only enzyme specific, but also species specific.

    A 3% solution of potassium permanganate is effective at acid pH as a fixative for the small noradrenaline-containing granular vesicles of the symapthetic nervous system. The observed high electron density of the central granules in the vesicles is probably due to precipitated MnO2 which is reduced KMnO4 in a coupled oxidation of noradrenaline to noradrenochrome. A 3% solution of KMnO4 in dichromate buffer (pH 6.0) is particularly suitable for the fixation of the vesicles. This may be due to the additional precipitation of a chromic compound reduced from the dichromate.

    Specific granules of the atypical spermatids in the pond snail were tested histochemically. There are large and small atypical sperm lines, both having these specific granules. These granules are morphologically similar and have been thought to be identical. Present study, however, reveals that they are different from each other in properties. They were equally positive to PAS and some general procedures for proteins. But, tryptophan was found only in the granules of the spermatids of small line, while tyrosine and potassium were observed in those of the large line. Definite acidity of these granules is not probable because of the negative reaction to colloidal iron. The granules of large line were stained with pyronin, Janus green B and alcian blue, but not with toluidine blue, methylene blue and neutral red. This suggests some physical relation of the dyes with the granules, rather than chemical reaction of the dyes. Complete lack of basophilia of the granules of small line may be attributed to some structural difference.

    The motor column of spinal cord in chick embryos after stage 25 becomes thick to form the lateral motor column in limb levels. Hofmann's nucleus is established in stage 34. The visceral motor cells originate from the motor column of the thoracal region. They cross dorsomedially over the arcuate fibers and the band of alkaline phosphatase at the dorsoventral boundary and finally settle at the dorsolateral angle of central canal. The lateral motor column and Hofmann's nucleus show an intense activity of cholinesterase but none of alkaline phosphatase. After stage 36, succinate dehydrogenase, adenosine triphosphatase and cytochrome oxidase tend to concentrate in them. Visceral motor cells, motor column of thoracal segment and medial motor column in limb levels are moderately positive in cholinesterase and alkaline phosphatase, but weak in succinate dehydrogenase, adenosine triphosphatase and cytochrome oxidase. Visceral motor center shows monoamine oxidase activity during stage 32-34. These suggest that the cells of lateral motor column and Hofmann's nucleus have a gradual shift towards aerobic state from stage 36, while the other groups of motor cells remain anaerobic.
