A method to detect and measure HB-Ab by means of radioimmunoassay using “Ausria-125” kit (Abbott) is proposed: That is, HB-Ab coated tube and I
125 labeled HB-Ab are used to test HB-Ab. This method is about 4, 000 times more sensitive than crossimmunoelectrophoretic method.
To test HB-Ab, aliquot amounts of HB-Ag which are about equivalent to HB-Ab coated in each tubes were added in the tubes; after washing the excess of HB-Ag, test solutions containing HB-Ab are added in the HB-Ag bound HB-Ab coated tubes; this is washed and then labeled HB-Ab is added so as to bind all of the remaining HB-Ag; after washing, radioactivity of I
125 bound in the tubes is counted by scintillation spectrometer.
This method was proved to be negative of the competitive reaction to antigen of antibodies which might be caused by the difference between HB-Ab of the test solution and labeled HB-Ab. Moreover, it was proved convenient to assay both HB-Ag and HB-Ab with same HB-Ab coated tubes and labeled HB-Ab and to compare both results directly. This method is recommended to apply for the routine test as a simple and sensitive tool for screening HB-Ab at high sensitivity.
