An α-linked D-manno-D-galactan, glycogen, and three kinds of branched (1 → 3)-β-D-glucans were isolated from
Fukurotake, the fruiting body of
Volvariella volvacea by successive extractions with cold and hot water, cold and hot alkali, and dimethyl sulfoxide. The mannogalactan, purified from the cold water extract, had a MW of 4 × 10
5, and consisted of an α-(1→6) linked D-galactose backbone, one out of every three D-galactose residues being substituted with a single α-D-mannosyl group. The glycogen, isolated from the hot water extract, had a MW of 12 × 10
5, and 14-15 D-glucose residues as an average chain length, as revealed by methylation analysis. The α-(1→4)-linked unit chains of this glycogen were distributed from DP 6 to 13, approximately in equal numbers.
There were three kinds of branched (1→3)-β-D-glucans, isolated from alkali and dimethyl sulfoxide extractions. They contain a backbone of (1→3)-liriked D-glucose residues with side chains of single D-glucosyl groups, but having different degrees of branching. In addition, alkali-extracted glucans contain small but significant proportions of (1→6)-linked sugar units.
Among these polysaccharides, the cold alkali-extracted glucan (degree of branching, 1:5) showed a potent antitumor activity against Sarcoma 180 solid tumors implanted in mice, and chemical modifications changed its original activity, confirming our previous results. Other polysaccharides, such as the mannogalactan and other (1→3)-β-D-glucans, showed no or lower antitumor activity.

A mesophilic and three thermophilic clostridial species were newly isolated as potent xylan utilizing bacteria from mesophilic and thermophilic methane sludge, respectively. The mesophilic strain, X-4, had a characteristic fermentation pattern of xylan, producing ethanol and acetic acid in addition to hydrogen and carbon dioxide. The thermophiles, XY-6, XY-10 and XY-23, were strains of the typical butyric acid fermentation type. The utilization of xylan by mesophilic strain X-4 was evident at late exponential or early stationary growth phase followed by an increase in production of xylanase in the culture fluid, while xylan was fermented by the thermophilic strains from the beginning of their cultivation. Cell-bound xylanase was detected in the three thermophilic strains. Enzyme secreted into the medium (supernatant xylanase) was detected in strains XY-6 and XY-10 but not in strain XY-23. The actions of xylanase preparations from X-4 and XY-6 were characterized, indicating that cell-bound β-xylosidases were also involved in the degradation of xylan by both strains.

Extracts of various flower tissues of tobacco with 70% methanol inhibited tobacco seed germination differently. Among them, extracts of stigma and anther were very inhibitory. When the extracts were partitioned between ethyl acetate and water, the activity of the ethyl acetate layer was stronger than that of the water layer. Stigmas and anthers had more abscisic acid (ABA) than the other floral tissues, which matched the results of the germination tests well. Guided by a bioassay using the inhibitory effects on tobacco seed germination, MeABA, ABA, and ABA-β-Dglucopyranoside were isolated and identified from stigmas. All of the MeABA isolated did not seem to be an artefact produced by esterification with the solvent, for MeABA was detected even when stigmas were extracted with other solvents than methanol. MeABA and ABA did not inhibit tobacco pollen germination and elongation
in vitro.

Excellent L-proline producers were screened for among sulfaguanidine resistant mutants derived from three typical L-glutamic acid-producing bacteria:
Brevibacterium flavum,
B. lactofermentum, and
C. glutamicum.
The best strain, No. 199, is a sulfaguanidine resistant mutant derived from an isoleucine auxotroph of
B. flavum 2247 by nitrosoguanidine. Strain No. 199 produced 35 mg/ml of L-proline after 72 hr of cultivation with 10% glucose as a carbon source. The strain also accumulated purine bases such as adenine, guanine, and hypoxanthine,
i.e., degradation products of purine nucleotides. In the mutant, 1.6-2.0 fold more intracellular ATP was found than that in the parent strain; it is a substrate of glutamate kinase relating to L-proline biosynthesis.
On the contrary, the levels of intracellular glutamic acid, a substrate of glutamate kinase, were similar among these strains.
It was confirmed that the increment of internal ATP, which was important in the L-proline production mechanism, was very effective in the improvement of L-proline producers.

Appreciable amounts of carotenoid in chum salmon were decomposed during spawning migration, although muscle carotenoid accumulated in the integument and ovaries. This decomposition of carotenoid was considered to occur in the liver tissue. The liver extracts with 0.5M KCl bleached astaxanthin in the presence of linoleic acid
in vitro. The astaxanthin bleaching activity was closely related to the size of the liver tissue. This activity in liver tissue was not completely lost by heating at 100°C for 10min. The liver extracts showed an absorption maximum at 410-415nm, along with two broad ones at 540-545 and 575-580 nm. These results suggested that carotenoids transported from muscle to liver by serum lipoproteins are decomposed by heme proteins of the liver tissue.

Human-human hybridomas were produced by fusion of a malignant 6-thioguanine resistant human B lymphoblastoid cell line and normal lymphocytes. The fusion efficiency with lymphocytes from peripheral blood was elevated by preculture of the lymphocytes in the presence of lipopolysaccharides. However, the preculture did not enhance the efficiency with lymphocytes from lung cancer tissue. The resulting hybridomas secreted mostly IgG and/or IgM. A small number of hybridomas secreted IgA. Two IgA secreting hybridomas, 9P-13-4 and 9L-27-9 also produced IgM-like immunoglobulin even after repeated cloning, indicating these hybridomas secrete IgA and IgM-like immunoglobulins simultaneously. The IgM-like immunoglobulin has a molecular weight of 900 K and contains 3 monomeric IgMs and 2 IgAs. This indicates that it is therefore possible to produce a IgM-like immunoglobulin binding to two different antigen determinants, by hybridoma technology.

The growth-promoting effects of a dialyzate of the extract from a thermophilic blue-green alga,
Synechococcus elongatus var. on several humal cell lines were studied in serum-free media. The dialyzate promoted the growth of RPMI 8226 (a myeloma cell line), Molt-4 (an acute T-lymphatic leukemia cell line), and HMY-2 and HO-323 (B-lymphoblastoid cell lines). The supplementation of basal RDF medium with the dialyzate enhanced the growth of HMY-2, HO-323, and RPMI 8226 cells more than that with a mixture of insulin, transferrin, ethanolamine, and selenite. Culture of RPMI 8226 cells for 2 weeks was achieved by use of the dialyzate-supplemented RDF medium. The growth-promoting activity of the dialyzate was heat-stable.

Besides its amide hydrolase activity, the amidase from
Brevibacterium sp. R312 also exhibits an acyl-transferase activity.
The mechanism of the transfer reaction of the acyl from acetamide to hydroxylamine was studied. This is a "Bi Bi Ping Pong" type reaction. The kinetic parameters of the reaction were determined:

N6-substituted adenine,
N6-substituted adenosine and 6-alkoxypurine with odd numbers of methylenes had relatively high activity, while the corresponding 2-methyl-4-substituted-aminopyrido[3, 4-
d]pyrimidines with even numbers of methylenes had high activity in an
Amaranthus betacyanin test and lettuce seed germination test. These results suggest that the ω-phenyl groups of the cytokinins play a specific role for cytokinin-receptor binding and that the pyrido[3, 4-
d]pyrimidines interact with a receptor in a different binding mode.

The isolation and partial characterization of a cadmium-binding protein from soybeans harvested in a cadmium-polluted field were performed. Sephadex G-75 chromatography of the cadmium fraction with an apparent molecular weight of 10, 000, which was released from the macromolecules in the presence of 2-mercaptoethanol, showed two components with apparent molecular weights of 22, 000 and 9200. The cadmium fraction gave a Coomassie-blue staining band at
Rf 0.92 and a weakly stained zone at about
Rf 0.6 on polyacrylamide gel electrophoresis. Amino acid composition analysis of the cadmium fraction revealed a low half-cystine content and high acidic amino acids contents. SDS-Polyacrylamide gel electrophoresis of the cadmium fraction gave a single band at a position corresponding to a molecular weight of 21, 000 and a broad band at one corresponding to about 9000. The cadmium component of a molecular weight of 21, 000 bound tightly to DEAE-Sephadex A-25 resin.

Bifidobacteria showed higher hydrolyzing activity toward various
p-nitrophenyl glycosides (
p-NP glycosides) than some other intestinal bacteria. The reactions commonly found in the organisms involved
p-NP β-D-galactoside,
p-NP α-D-glucoside and
p-NP β-D-fucoside. Analysis of the enzyme species suggested that the β-D-fucoside-hydrolyzing reaction, which is undetectable in other bacteria, was catalyzed by β-D-galactosidase in many bifidobacteria and by β-D-glucosidase in some strains.
β-D-Galactosidase, which hydro lyzed
p-NP β-D-fucoside (with 12% of its reactivity to
p-NP β-D-galactoside), was purified to homogeneity from
Bifidobacterium longum 401. The enzyme was distinct from other bacterial β-D-galactosidases in its higher activity toward lactulose than lactose and the insensitivity of its formation to the carbon source in the culture medium. Some properties of the β-D-galactosidase are described and compared with those of the lactase from the same organism.

Two β-D-glucosidases were purified to homogeneity from
Bifidobacterium breve 203: one β-D-glucosidase I; molecular weight, 96, 000) showed reactivity toward
p-nitrophenyl (
p-NP) β-D-fucoside, 74% of that to
p-NP β-D-glucoside, and the other (β-D-glucosidase II; molecular weight, 450, 000) did not. They also differed in their thermal and pH stabilities. Laminaribiose, cellobiose and gentiobiose were hydrolyzed by β-D-glucosidase I, with 53%, 34% and 3% of the reactivity in the case of
p-NP β-D-glucoside, and by β-D-glucosidase II, with 53%, 6% and 107% of the reactivity. The reaction of β-D-glucosidase I with
p-NP β-D-fucoside was enhanced by the addition of glucose and other monosaccharides to the reaction mixture, whereas that with
p-NP β-D-glucoside was not affected. The activity of β-D-glucosidase II with
p-NP β-D-glucoside was inhibited by glucose.

The use of desorption chemical ionization (D/CI) mass spectrometry for the structural characterization of the abscisic acid (ABA)-conjugated metabolites, abscisic acid-β-D-glucosyl ester (ABAGE), 4'-
O-dihydrophaseic acid-β-D-glucoside (DPAGS) and β-hydroxy-β-methylglutarylhydroxyabscisic acid (HMG-HOABA), was studied. The effects of the D/CI source temperature on the spectral features are described. Enhanced quasi-molecular ion intensities (
e.g., [M•NH
4]
+) were observed in the D/CI(NH
3) spectrum at a low source temperature (170°C). When a higher source temperature (250°C) was used, more extensive fragmentation occurred, and structurally diagnostic fragment ions appeared in the spectra. The spectra obtained at two degrees of source temperature were complementary to each other for the structural characterization.

Fourteen strains of the genera
Rhodococcus,
Nocardia, and
Corynebacterium were found to produce substances which flocculate kaolin clay.
Rhodococcus erythropolis produces a kind of the microbial flocculant. It had very interesting and unique flocculating characteristics. It could efficiently flocculate all suspended solids in aqueous solution tested. Among those effectively tested were microorganisms such as
E. coli and alcohol yeast, activated sludge,
Microcystis aeruginosa (AOKO), kaolin clay, muddy water, river dredging muddy water, river bottom sediment (HEDORO), ash from a steam-power station, and chacoal. These results indicated that the flocculant from
R. erythropolis had a wide flocculating activity against both organic and inorganic materials. Cationic ions such as Ca
2+ also significantly increased the flocculating activity.

Addition of yeast extract and casamino acids stimulated the production of a flocculant by
Rhodococcus erythropolis. The favorable substrates for flocculant formation were fructose (or glucose), urea, and yeast extract. Optimum culture temperature and optimum culture pH for the production of the flocculant were 30°C and pH 8.5-9.5, respectively. Excess aeration decreased the production of the flocculant. The production of the flocculant paralled the growth curve. More than 90% of the flocculating activity was in the culture broth and less than 10% in the cells.

To find the mechanism of the amino-carbonyl reaction of proteins with reducing sugars, we examined the reactivity of
Nα-acetyl-tryptophan (N-Ac-Trp), a model of tryptophan residues, with various sorts of carbonyl compounds, formaldehyde, acetaldehyde, glyoxal, methylglyoxal, butanedione, and acetone, on incubation at pH 6.8 and 50°C. Aldehydes reacted with N-Ac-Trp, but ketones did not.
The most abundant reaction product between N-Ac-Trp and glyoxal (Trp-GO-1) was identified as
Nα-acetyl-1-(2-hydroxy-1-oxoethyl)-L-tryptophan and that between N-Ac-Trp and methylglyoxal (Trp-MG-1) was identified as
Nα-acetyl-1-(1-hydroxy-2-oxopropyl)-L-tryptophan. From the UV spectrum, a major reaction product between N-Ac-Trp and 3-deoxyglucosone (Trp3DG-1) was supposed to be analogous to Trp-MG-1.
While Trp-GO-1 polymerized itself very little, Trp-MG-1 did produce some polymers. This suggests that Trp-MG-1 and Trp-3DG-l may participate in the crosslinking of proteins.

Four trypsin inhibitors were isolated from seeds of okra (
Abelmoschus esculentus L.) and named okra trypsin inhibitors (OTI) A, B, C, and D. The molecular weights of all the inhibitors were estimated to be about 20, 000 by gel filtration on Sephadex G-100 and SDS-polyacrylamide gel electrophoresis. All of these inhibitors inhibited bovine trypsin strongly with the formation of complexes at a 1:1 molar ratio and three (OTI-A, OTI-B, and OTI-C) of them also weakly inhibited bovine chymotrypsin, but not simultaneously. Subtilisin Carlsberg, pronase, papain, and porcine pepsin were not inhibited by these inhibitors. The trypsin inhibitory activity of all four inhibitors was suppressed by treatment with 1, 2-cyclohexanedione but not with 2, 4, 6-trinitrobenzenesulfonate, suggesting that each of the inhibitors contains an arginine residue at the reactive site.

The hypocholesterolemic efficacy of various polyunsaturated fatty acids was compared in rats given cholesterol-enriched diets with (0.004%) or without indomethacin, the cyclooxygenase inhibitor. Evening primrose oil (EPO, linoleic+γ-linolenic), safflower oil (SFO, linoleic) or soybean oil (SBO, linoleic +α-linolenic) were added to diets at the 10% level. The serum cholesterol level of the EPO group was consistently lower than the other groups and after 3 weeks, it was significantly different from the SFO group without indomethacin and the SBO group with indomethacin. In rats fed EPO, the aorta tended to produce more prostacyclin whereas the concentration of plasma thromboxane B
2 was much lower than in rats fed SFO or SBO. The effects of indomethacin on these eicosanoids were less evident in rats fed EPO. Thus, in addition to the hypocholesterolemic action of γ-linolenic acid (GLA) in preference to linoleic and possibly α-linolenic acid, GLA appears to cultivate an environment suitable for the prevention of carbiovascular disease even in the presence of excess cholesterol in the diet.

N-α-substituted benzyl benzenesulfonamides were synthesized and their phytotoxic activities against barnyardgrass and rice plants were tested. Most of the compounds exhibited potent phytotoxicity against barnyardgrass, and high selectivity between transplanted rice seedlings and barnyardgrass. β-
N-Acetyl-D-glucosaminidase (EC 3.2.1.30) was purified from the alimentary canal of the silkworm,
Bombyx mori, by ammonium sulfate fractionation and chromatography with hydroxylapatite, DEAE Bio-Gel A, chromatofocusing, and Sephacryl S-200. The purified enzyme was a single band on disc-PAGE. The molecular weight was 119, 000 by gel filtration and 125, 000 by SDS-PAGE. The enzyme was separated into two peptides whose apparent molecular weights were 67, 500 and 57, 500 by SDS-PAGE. The pi was 4.86 by chromatofocusing. The optimum pH was 5.5 to 6.0 and the optimum temperature, 45°C, using pNp-β-GlcNAc as the substrate. The enzyme was stable from pH 5.5 to 8.5 and below 30°C. It was strongly inhibited by HgCl
2. Small
N-acetylchitooligomers were as good substrates as pNp-β-GlcNAc, and the enzyme cleaved colloidal chitin to GlcNAc, even though the relative velocity was slow. Smaller

-1. The enzyme precipitated in as band with moulting fluid chitobiase antiserum, but not with haemolymph exo-β-N-acetylglucosaminidase antiserum. The results suggest that this enzyme is an exo-type enzyme which is involved in the degradation of chitin to GlcNAc.

Asymmetric hydrolysis of (
dl)-1-acyloxy-2-halo-1-phenylefhanes by lipoprotein lipase Amano P from
Pseudomonas fluorescens and the lipase from
Chromobacterium viscosum afforded the optically active (
R) residual substrates and (
S)-2-halo-1-hydroxy-1-phenylethanes in 100% enantiomeric excess (
e.e.). The length of acyl residues from acetyl to octanoyl in the substrates did not influence the enantioselectivity.
Both enantiomers of optically active styrene oxides were synthesized from the enzymatic products.

Fragment peptides of milk proteins containing Tyr-X-Phe or Tyr-X

2-Phe were synthesized and their opioid activities were evaluated by the guinea pig ileum and radioreceptor assays. Among the peptides tested, the human β-casein(51 - 54) amide, Tyr-Pro-Phe-Val-NH2 (valmuceptin) was the most active, more than the bovine counterpart, morphiceptin. The human β-casein(41 - 44) amide, Tyr-Pro-Ser-Phe-NH2, and the α-lactalbumin (50-53) amide, Tyr-Gly-Leu-Phe-NH2, were also active in the guinea pig ileum assay. These were named β-casorphin and α-lactorphin, respectively. All of these peptides were not as active in the mouse and rabbit vas deferens assays.
