Simple and partial correlations were computed among 23 physico-chemical characteristics of
L. dorsi muscle from lambs. The object was to single out those correlations which are inherent in the muscle composition and those which are dependent on nutritional status and age of the lambs. A large number of simple correlations between different characteristics were found to be significant.
However, the partial correlations, adjusted for plane of nutrition and age, revealed only for a small number of significant relationships which were inherent in muscle composition.
Most of the correlations were spurious, in the sense, that they depended mainly on the plane of nutrition and age of the lambs.
The intensity of correlation improved between some muscle characteristics by eliminating the effect of plane of nutrition and age. This suggests that the variation in those characteristics was not only due to plane of nutrition and age but some third factor was also involved in affecting their relationships.

The breakdown of cellular protein was investigated in
Bacillus subtilis labeled with glycine-2-
3H or L-phenylalanine-U-
14C at different stages of vegetative growth and sporulation. In cells labeled with L-phenylalanine-U-
14C, multiple protein turnover was observed. However, in cells labeled with glycine-2-
3H, the patterns of protein turnover were quite different in the stages of growth and sporulation; proteins which were labeled at the early stationary phase were degraded rapidly, but those labeled at the late sporulation stage were hardly degraded. It was found that glycine incorporated into cells at the late sporulation stage was mainly utilized for biosynthesis of the spore coat protein. These data suggest that the spore coat protein which contains relatively large amounts of glycine is little subject to further degradation.

Streptomyces orientalis was selected as a microorganism which produced chitinase having lytic activity on
Rhizopus cell wall.
Two chitinases (chitinase I and II) which had very similar properties were purified to an ultracentrifugally homogeneous state from the culture broth of this organism by SP-Sephadex and Sephadex G-100 chromatographies.
Molecular weights of chitinase I and II were estimated as about 33, 000 and 25, 000, and isoelectric points as pH 8.80 and 8.65, respectively. Both chitinases were most active at pH values between 5.5 and 6.5, and were stable in the range of pH 6.0 to 8.0 at 40°C for 3 hr. They were stable at temperatures below 50°C for 15min.
Rhizopus cell wall was almost completely degraded by a cooperative action of these chitinases, and a protease and a chitosanase from
Bacillus R-4.

An aminopeptidase active on L-Val-L-Val-L-Val-L-Ala was purified from rabbit skeletal muscle by the method including ammonium sulfate precipitation, DEAE-cellulose chromatography, gel-filtration on Sephadex G-200, rechromatography on DEAE-cellulose, hydroxylapatite chromatography and rechromatography on Sephadex G-200. Polyacrylamide gel disc electrophoresis showed that the enzyme thus obtained was homogeneous. The specific activity of the purified enzyme was 1500 times that of the original muscle extract. The enzyme had an optimal pH in a range of 6.0_??_7.6 and was stable in pH 6.1_??_8.1. Molecular weight of the enzyme was estimated to be 160, 000 from the result of gel-filtration on Sephadex G-200. The enzyme showed specificity for tri-, tetra-, penta-, and hexapeptides. The analytical data of liberated amino acids showed that the enzyme was an aminopeptidase active on these oligopeptides. The enzyme was strongly inhibited by
N-ethyl-maleimide and EDTA.

Hydroxyproline tri- and tetra-arabinosides were isolated from suspension-cultured tobacco cell wall preparation by Ba(OH)
2 treatment followed by gel filtration and ion-exchange chromatography. Methylation analysis showed that they consisted of terminal, 1, 2-linked and 1, 3-linked arabinofuranosyl residues in molar ratios of 1:2:0 and 1:2:1, respectively. A part of
trans-4-hydroxy-L-proline in the arabinosides was revealed to have been isomerized into
cis-4-hydroxy-D-proline during the alkaline hydrolysis.

Evidence of the presence of
trans-2-hexenol among tomato volatile components was shown using heavy solvent extraction and adsorption-desorption techniques.
The action of alcohol deshydrogenase isolated from tomato fruit on
trans-2-hexenal shows that the pathway for the formation of
trans-2-hexenol is enzymatic. The presence of hexanal and hexanol besides this alcohol indicate that the reduction of the double bonds by the enzymatic preparation is also possible.

Phospholipids of
Gluconobacter cerinus were investigated. The cells of this bacterium were found to have cardiolipin, phosphatidylglycerol, phosphatidylethanolamine and phosphatidylcholine together with three ornithine-containing lipids. Phosphatidylcholine was the most abundant of these phospholipids, and calculated at 31.0% of total phospholipids in the cells at the late logarithmic phase. The fatty acid of this lipid was composed of palmitate, stearate and vaccenate, and there were no remarkable differences in quality among fatty acids of these four phospholipids. The phosphatidylcholine was also found in other acetic acid bacteria.

The interaction between ovomucin and egg white proteins has been investigated. Ovomucin interacted not only with lysozyme but also with such proteins in egg white as ovalbumin and conalbumin. The interaction increased correspondingly as the lysine content of proteins become higher. The maximum turbidity of ovomucin-lysozyme aggregates was proportionally dependent on the rate of acetylation of lysozyme, but that of ovomucin-ovalbumin aggregates was slightly dependent on the rate of acetylation of ovalbumin. The ovomucin-protein interaction decreased remarkably by the removal of sialic acid in ovomucin, but increased by the removal of polyhydroxyl group in sialic acid. From these results, the binding site of interaction was considered in detail.

A comparative study of the effects of processing on the carotenoid composition of the two popular varieties of tea namely,
Thea sinensis and
Thea assamica has been carried out. The results show that the carotenoid pattern in tea dust is very similar to that in tea leaves in spite of the drastic manufacturing conditions. However, about 20% of the carotenoids are found to be lost during manufacture. Also, the carotenoid loss during tea preparation has been carried out. The results reveal that about 10% of the carotenoids are lost from the tea dust during tea brewing.

The determination of dissolved hydrogen concentration (C
H2) in the culture medium of hydrogen bacteria was performed. Dissolved hydrogen in the medium was stripped by bubbling nitrogen and thus stripped hydrogen was quantitatively determined by a gas chromatograph. C

In cell-free homogenates of
Saccharomyces cerevisiae, Denmert (S-1358) inhibited the incorporation of radioactivity from DL-mevalonate-2-
14C into 14-desmethyl-lanosterol, 4α-methyl-cholesta-8, 24-dien-3-one, 4α-methyl-zymosterol and 4-desmethyl sterols (zymosterol and episterol) at a concentration of 10
-4M. Concomitantly, a large accumulation of radioactivity was observed in the lanosterol fraction.
In good agreement with the results described above, Denmert inhibited the conversion of
14C-labeled lanosterol to 4-desmethyl sterols, while the conversion of
14C-labeled 14-desmethyl-lanosterol to 4-desmethyl sterols was hardly affected by the fungicide. It is therefore evident that Denmert is a potent selective inhibitor of the demethylation at the C-14 position in ergosterol biosynthesis.
The fungicide, triarimol, was also found to exhibit the same effect on sterol biosynthesis as Denmert.

A method for the conversion of allyl alcohol into glyceraldehyde derivatives by the cooxidation with thiol by oxygen and the successive Pummerer reaction is described. Several methods to transform β-hydroxy or β-oxo sulfoxides into “activated ester forms” of α-hydroxy acids are reported. By these methods, derivatives of mandelic acid and lactic acid were prepared from 2-hydroxy-2-phenylethyl
p-tolyl sulfoxide and phenylsulfinylacetone, respectively. From a derivative of β-alanine were prepared some derivatives of 4-amino-2-hydroxy-butyric acid which is used for the modification of aminoglycoside antibiotics.

The soluble ovomucin obtained from the liquid part of thick white by gel filtration on a Sepharose 4 B was an aggregated and polymerized molecule (intrinsic viscosity was 365ml/g and molecular weight was 8.3×10
6) and it was unable to dissociate the soluble ovomucin into two components without modifications.
Molecular weight and reduced viscosity of the soluble ovomucin decreased markedly with time of sonication. By the sonication for 10min, it was successful to fractionate it into carbohydrate rich and poor component by density gradient electrophoresis, cellulose acetate electrophoresis and DEAE-cellulose column chromatography.
Concerning carbohydrate and amino acid compositions of two components obtained from the sonicated soluble ovomucin, it was found that the carbohydrate poor component corresponded to the reduced S-component or the reduced α-ovomucin, and the carbohydrate rich component to the reduced F-component or the reduced β-ovomucin.
It was considered that the sonicated soluble ovomucin was an intermediate of the aggregated, polymerized ovomucin (the soluble ovomucin) and the monomeric ovomucin (the sonicated and reduced soluble ovomucin).

The association and conformational change of α
s1-casein by an increase of ionic strength were investigated.
s20, w was increased rapidly by addition of NaCl and reached to constant above 0.2M NaCl. It was shown by gel filtration on Sephadex G-100 that dimer of α
sl-casein (at zero m NaCl) changed to tetramer (above 0.2M NaCl). This association proceeded with the burying of tyrosyl and tryptophyl residues, and caused the decrease of intrinsic viscosity. α
s1Casein was speculated to approach to a spherical shape when it changed from the dimer to tetramer. However, no change of the secondary structure was caused by this association.
The formation of aggregates of α
s1-casein by calcium was decreased rapidly up to 0.2M NaCl. This change was parallel to the formation of tetramers, and the gel filtration pattern of α
s1-casein was not changed at 0.3M NaCl by addition of 6mM CaCl
2. Thus, it appears that the formation of tetramers is responsible for the inhibition of the formation of aggregates of the protein by calcium.

A proteinaceous inhibitor of auxin-induced ethylene synthesis by mungbean hypocotyls was purified from mungbean seeds by chromatography on CM-Sephadex C-50 and gel filtration on Sephadex G-200. The molecular weight was estimated to be about 89, 000 by gel filtration on Sephadex G-200. The inhibitory protein consisted of two subunits with molecular weights of about 58, 000 and 31, 000.

A procedure is described for purification of phosphoglucomutase [EC 2. 7. 5. 1] from beef liver. The purified enzyme preparation was homogeneous on the analysis of ultracentrifugation and zone electrophoresis. The molecular weight was determined to be 64, 000 by the meniscus depletion method.
The amino acid composition of liver phosphoglucomutase was very similar to that of the rabbit muscle enzyme.
The reaction mechanism of liver phosphoglucomutase was examined kinetically. The results of kinetical experiments strongly suggested that the reaction of liver phosphogluco-mutase proceeds
via “ping-pong” mechanism.
Liver phosphoglucomutase activity was remarkably inhibited by Fru-1, 6-P
2, glycerate-2, 3-P
2 and glycerate-1, 3-P
2. Role of the bisphosphate compounds on the control of carbohydrate metabolism is discussed.

Liver phosphoglucomutase was found to catalyze also the reaction of Glc-1, 6-P
2 formation from Glc-1-P and Fru-1, 6-P
2 or Glc-1-P and glycerate-1, 3-P
2. The specific activity of Glc-1, 6-P
2 formation from Glc-1-P and Fru-1, 6-P
2 was 1/9200 of that of the mutase activity. The activity of Glc-1, 6-P
2 formation from Glc-1-P and glycerate-1, 3-P
2 was 1/122, 000 of the mutase activity. From the results of the kinetics and the thermal inactivation experiments, the reaction of the mutase and Glc-1, 6-P
2 synthesis were strongly suggested to occur at the same active site of liver phosphoglucomutase.
Liver phosphoglucomutase exhibited the Glc-1, 6-P
2 phosphatase activity only in the presence of xylose 1-phosphate. The specific activity of phosphatase was only 1/154, 000 of that of the mutase activity.

Four kinds of the enzyme reactions have been reported for the synthesis of Glc-1, 6-P
2. However, any activity of Glc-1-P dismutase and phosphoglucokinase was not observed in the beef liver homogenate. When the liver homogenate was incubated with Glc-1-P and Fru-1, 6-P
2, a significant amount of Glc-1, 6-P
2 was formed. The Glc-1, 6-P
2 synthesis activity from Glc-1-P and Fru-1, 6-P
2 was caused by the action of phosphoglucomutase present in the liver homogenate. The most remarkable activity for Glc-1, 6-P
2 synthesis was observed when the homogenate was incubated with Glc-1-P and glycerate-1, 3-P
2. The Glc-1, 6-P
2 synthesis activity from Glc-1-P and glycerate-1, 3-P
2 was separated from the major peak of phosphoglucomutase activity by DEAE-Sephadex chromatography. The peak of Glc-1, 6-P
2 synthesis activity, however, still retained phosphoglucomutase activity.
Glc-1, 6-P
2 phosphatase activity was mainly observed in the mitochondria and microsome fraction. The properties of Glc-1, 6-P
2 phosphatase were differentiated from those of acid phosphatase and Glc-6-P phosphatase.

The myosin total rod, which consists of smaller segments of light meromyosin and heavy meromyosin subfragment-2 (HMM S-2), prepared by limited papain digestion of rabbit myosin, was purified by Sepharose-2 B column chromatography. The purified total rod was more homogeneous than any previously reported, and the sodium dodecyl sulfate (SDS) gel electrophoretic method yielded a molecular weight of 22-23×10
4 (11-11.5×10
4×2).
Transition temperatures of this purified myosin total rod obtained from the melting profile during heating were 47.5 and 55°C. The results of ORD and CD measurements showed almost full reversibility upon cooling after thermal treatment. However, the results obtained from difference spectra and fluorescence spectra showed incomplete reversibility with hysteresis.
This ostensible dichotomy concerning the structural thermostability of the rod portion of myosin molecule may mean that although ORD and CD studies show almost full reversibility of the helix-coil transition, local irreversible conformational changes, involving aromatic amino acid residues take place. This fact suggests that the renatured α-rope of the myosin total rod can exhibit different properties than the native molecule under conditions where no discernible loss in helix content occurs.

Five new metabolites related to IAA were isolated from aqueous methanol extract of rice bran and their structures were elucidated by spectroscopic methods to be 5-hydroxydioxindole-3-acetic acid (Ia), methyl ester of la (Ib), methyl dioxindole-3-acetate (IIb), methyl 5-hydroxyoxindole-3-acetate (IIIb) and methyl oxindole-3-acetate (IVb), respectively. The presence of these metabolites which are likely to be intermediates of oxidation of IAA to β-acid suggests such oxidation pathway operating in rice seed.
