The heat denaturation process of myosin and the effect of the modification of amino groups of myosin with β-naphthoquinone-4-sulfonic acid (NQS) on heat denaturation reaction were studied by Using SDS polyacrylamide gel electrophoresis.
Heavy chain and one of the light chains, g
2 disappeared in the supernatant after centrifugation of heat treated myosin during two hours'heating, whereas two sorts of the light chains, g
1 and g
3 ere still found in the supernatant. In adition, a new peptide fraction appeared during heating.
Heat denaturation of myosin was significantly suppressed by the modification of amino groups of myosin with NQS, where only heavy chain's electrophoretic pattern was influenced by NQS-modification.

The effect of xylan from beech (
Fagus crenata Blume) sapwood on the plasma and liver cholesterol levels and on the excretion of cholesterol and bile acid in feces were studied in rats fed hypercholesterolemic diet. Supplementation with 5% of xylan to the hypercholesterolemic diet for one week caused about 26% and 17% of depression in the plasma and liver cholesterol levels, respectively, compared with the control group and when feeding period was continued two weeks they showed about 28% and 37% of depression. The intestinal absorption of cholesterol and bile acid was significantly inhibited in the groups fed xylan. Moreover, when the hypercholesterolemic diet was replaced by the test (xylan) diet, the accumulated liver cholesterol level was lowered more than that of normal group. This finding is seemed to be due to increased catabolism of cholesterol to bile acid by continued removal of bile acid.
On the basis of these results, it can be possibly concluded that the most important hypocholesterolemic effect of xylan is due to suppression of the intestinal absorption of cholesterol and bile acid.

The conditions of promotion of autolysis of three strains of lactobacilli were investigated. The autolysis of
L. acidophilus, L. helveticus and
L. casei at exponential phase was remarkably enhanced by freezing storage at -20°C overnight.
The turbidity decrease of
L. acidophilus' cell suspension corresponds to the increase of cell free nitrogen compounds, glucosamine and DNA component. All these compounds were more rapidly released from the cells stored at -20°C than those stored at 3°C. The cells which were harvested at the exponential phase had higher autolytic activity than those at stationary phase. The storage of the cells at -20°C for 2 days or more could effectively promote the autolysis.
The activity was increased by Ca
2+ or Mg
2+. Optimum pH of the autolytic enzyme of
L. acidophilus was 6_??_7.

Adenylyl (5', 2') -adenosine 5'-phosphate ((2'-5') pA-A) was detected in crude crystals of 5'-AMP prepared from
Penicillium nuclease (nuclease P
1) digest of a technical grade yeast RNA. While (3'-5') A-A was split by nuclease P
1, spleen phosphodiesterase, snake venom phosphodiesterase or alkali, (2'-5') A-A was not split by a usual level of nuclease P
1 or spleen phosphodiesterase. Nuclease P
1 digests of 12 preparations of technical grade yeast RNA tested were confirmed to contain (2'-5') pA-A. Its content was about 1 to 2% of the AMP component of each RNA preparation. As poly (A) was degraded completely by the
Penicillium enzyme into 5'-AMP without formation of any appreciable amount of (2'-5') pA-A, the technical grade RNA is supposed to contain 2'-5' phosphodiester linkages in addition to 3'-5' major linkages.

Sixteen 2'-5' dinucleotides; (2'-5') pA-A, pA-G, pA-C, pA-U, pG-A, pG-G, pG-C, pG-U, pC-A, pC-G, pC-C, pC-U, pU-A, pU-G, pU-C, and pU-U were detected in nuclease P
1 digest of a technical grade yeast RNA by means of gel filtration on Sephadex G-10, DEAE-Sephadex A-25 column chromatography in the presence of 7M urea, paper electrophoresis and paper chromatography. Content of each dinucleotide was about 0.1 to 0.6% of the digest. As the sixteen 2'-5' dinucleotides were found in all of the digests of technical grade RNA preparations tested, each polynucleotide chain in the preparations may be concluded to contain several per cent of the 2'-5' minor phosphodiester linkages in addition to the 3'-5' major phosphodiester linkages.

Cell protein isolates were prepared from yeast (
S. cerevisiae) by alkali-extraction followed by acid precipitation. The relationships between alkali-treatment and nucleic acid contents in cell protein isolates were examined.
The isolate which was precipitated at pH 4.5 following extraction with 0.20 N NaOH at 80°C contained small amounts (less than 1% of the isolate) of nucleic acids. However, the content of nucleotides in the isolate which was precipitated at pH 4.5 following extraction with 0.20 N NaOH at 37°C was 9.13% of the product. Treatment by washing or dialysis of the isolate had little effect in removing the nucleotides in the isolate.
This finding was explained by the interaction of nucleotide to cell protein isolate. The binding energy was measured by Hummel's method.

During our studies on fermentative production of CDP-choline by dried cells of yeasts, CMP kinase system was found to be released into supernatant of the reaction mixture, while pyrophosphorylase remained bound to cells. The supernatant phosphorylated CMP to CDP and further to CTP. The phosphorylation was inhibited by glucose. However, the inhibition was reversed by addition of small amount of ATP. Addition of inorganic orthophosphate accelerated the phosphorylation by the supernatants of early stages of the reaction, for inorganic phosphate was deficient in the supernatants.

5-Fluorotryptophan (5 FT
*) -resistant mutants induced by one step mutation from a glutamate producing bacteria,
Brevibacterium flavum No. 2247, accumulated only 0.4g/liter of L-tryptophan at maximum. However, mutants resistant to higher concentrations of 5 FT, which were further induced from the 5 FT-resistant mutants, accumulated 1.4 to 2.4g/liter of L-tryptophan. Their anthranilate synthetases of the tryptophan-specific pathway were less sensitive to the feedback inhibition by tryptophan than the parent enzymes and were not repressed by tryptophan. Moreover, the relationship between the L-tryptophan accumulation and the resistance of the enzymes to the feedback inhibition was parallel to a considerable extent. 3-Fluorophenylalanine (3 FP) -resistant mutants induced from the 5 FT-resistant mutants produced about twice as much L-tryptophan as the parental strains. Their L-tryptophan accumulation was not affected by the addition of excess L-phenylalanine and L-tyrosine, in contrast to that in the parents. Their3-deoxy-D-
arabino-heptulosonate-7-phosphate (DAHP) synthetases of the common pathway were insensitive to the synergistic inhibition by phenylalanine plus tyrosine. Since mutants resistant to 5 FT and 3 FP accumulated L-phenylalanine and L-tyrosine in addition to L-tryptophan, phenylalanine auxotrophs were induced, which produced 6.2g/liter of L-tryptophan at maximum. The increment of the accumulation was almost equivalent to the amount of L-phenylalanine accumulated in the parental strain. These results were discussed on the basis of the regulatory mechanism for the aromatic amino acid biosynthesis in
B. flavum.

The effect of feeding 33mg of retinol daily for two days on liver and plasma lipids of rats has been studied. The secretion of liver TG into plasma of retinol fed rats has been measured by the use of palmitic acid-1-
14C and of Triton WR 1339. Liver and plasma lipids, TG, phospholipids and PC were significantly higher in retinol fed rats as compared to control rats. The incorporation of palmitic acid-1-
14C into liver TG, PC and PE was significantly higher in retinol fed rats. The labelling pattern with time of liver and plasma TG and PC shows that the secretion of liver TG and PC into plasma was impaired in retinol fed rats.

A correlation existed between the productivity of sclerin (SCL) and formation of sclerotia or dark pigmentation of old hyphae in
Sclerotinia libertiana Fuckel and
S. sclerotiorum de Bary. SCL stimulated the activities of phenoloxidase and peroxidase in a mycelial extract of
S. libertiana, which appeared to be involved in the maturation and pigmentation of sclerotia. On the other hand,
p-aminobenzoic acid (PABA), an inhibitor of phenoloxidase, inhibited the production of both SCL and sclerotia, where assimilation of PABA probably interrupted the biosynthetic process of SCL. When SCL was added at a high concentration, 4×10
-3M, to Czapek's agar medium a strain of
S. libertiana which originally formed sclerotia and a strain of
S. sclerotiorum which did not form sclerotia but produced melanin-like pigments at the edge of colony formed numerous sclerotium initials, which did not develop normal sclerotia. It was suggested that SCL played a role on induction of sclerotial formation and melanogenesis in these strains, in which a regulation of carbohydrate metabolism was involved.

Experiments using rats force-fed a protein-free diet supplemented with methionine, leucine, threonine, cystine or sulfur-containing compounds were conducted to investigate the effects of sulfur-containing amino acids on fat content in the liver and on the excretion of urinary nitrogen. The addition of methionine to a protein-free diet produced fat accumulation in the liver and reduced the excretion of urinary nitrogen, but that of leucine or threonine did not affect them. The addition of methionine to the protein-free diet produced a remarkable decrease in the concentration of threonine in the liver along with the increase in the concentration of methionine. On the other hand, the further addition of leucine to the mehtionine supplemented diet apparently eliminated the nitrogen sparing action of methionine and the effect of the added methionine on the concentration of threonine in the liver, but did not bring about any changes in the fat accumulation. The addition of cystine demonstrated the effects similar to that of methionine on the fat content in the liver and the excretion of urinary nitrogen, and showed a clear increase in the concentration of methionine and a great decrease in that of threonine, without changes in that of cystine, in the liver. However, fat accumulation of rats given the protein-free diet supplemented with cystine was not eliminated with the further addition of threonine to this diet. The addition of neither cysteic acid nor thioglycerol affected the free-amino acid concentration in the liver, but the latter produced the fat accumulation in the liver.

2-(2'-Hydroxy-2', 2'-diphenylethyl)-8-hydroxyquinoline was prepared via Grignard reaction involving the activated methyl group in position 2. This compound inhibited the action of the phenol oxidase prepared from prepupae of housefly. In a dipping test of the final instar larvae of housefly, it showed some inhibitory effects on the metamorphosis.

Sophoradin (I) [2', 4, 4'-trihydroxy-3, 3', 5-tris (3-methyl-2-butenyl) chalcone] which had been isolated from “Guang-Dou-Gen” (the root of
Sophora subprostrata CHUN et T. CHEN) was synthesized through Claisen rearrangement. The reaction of
p-hydroxybenzaldehyde and 3-chloro-3-methyl-1-butyne (III) gave 4- (1, 1-dimethylpropargyloxy) benzaldehyde (VIII), which was catalytically hydrogenated over Lindlar catalyst to afford 4- (1, 1-dimethylallyloxy) benzaldehyde (IX). IX was converted to 4-hydroxy-3-(3-methyl-2-butenyl) benzaldehyde (X) by Claisen rearrangement. The reaction of X and III gave 3-(3-methyl-2-butenyl)-4-(1, 1-dimethylpropargyloxy) benzaldehyde (XI). Condensation of 2-hydroxy-4- (1, 1-dimethylpropargyloxy) acetophenone (IV) and XI in alkaline solution gave a chalcone (XIII), which was catalytically hydrogenated over Lindlar catalyst to give 2'-hydroxy-4, 4'-bis (1, 1-dimethylallyloxy)-3-(3-methyl-2-butenyl) chalcone (XIV). XIV was converted to I by Claisen rearrangement.

The cleavage of double-stranded DNA by S1 endonuclease was studied by sucrose density gradient centrifugation analysis. The enzyme introduced no single-strand breaks into native T 7 DNA under conditions where heat-denatured T 7 DNA was completely degraded. By using enzyme at about 6 times higher the amount required for complete degradation of the heat-denatured DNA, it was possible to make a few single-strand breaks in native T 7 DNA. Under the conditions where native T 7 DNA is absolutely resistant to the enzyme, the susceptibility of locally altered structures naturally present and/or artificially induced in native double-stranded DNA to the enzyme was studied. It was evidenced that S1 endonuclease can cleave circular covalently closed, superhelical fl RFI DNA, depurinated T 7 DNA, bleomycin-treated T 7 DNA containing internal single-strand breaks, but not cleave intercalating drug-bound T 7 DNA.

6-Methoxy-2-benzoxazolinone has been synthesized by a simplified, improved procedure, the formation and reduction of 5-methoxy-2-nitrophenol and subsequent fusion of the corresponding amine hydrochloride with urea. In related work, 4-methoxy-2-(methoxymethoxy)-nitrobenzene was prepared.

Certain methionine auxotrophs of
Arthrobacter paraffineus and Bacillus species produce large amounts of O-acetylhomoserine (OAH). The methionine requirement of these auxotrophs could be satisfied by either cystathionine or homocysteine but not by homoserine. The cell-free extacts from the auxotrophs were found to be deficient in cystathionine γ-synthase activity. OAH and O-succinylhomoserine (OSH) could replace methionine in the auxotrophs which are deficient in homoserine-O-transacetylase. A methionine auxotroph of
Corynebacterium glutamicum also produced OAH, and the blocked step in the auxotroph appeared to be between cystathionine and homocysteine.
Cell-free extracts of
A. paraffineus, C. glutamicum and
Bacillus species catalyzed the formation of OAH from acetyl-CoA and homoserine, while a corresponding reaction with succinyl-CoA was not detected. Cystathionine γ-synthases in extracts of
C. glutamicum and
Bacillus species were specific for OAH, while the enzyme in extract of
A. paraffineus was rather specific for OSH though it reacted with OAH to a certain extent.
These results indicate that the biosynthesis of L-methionine in these bacteria involves OAH.

High phytin containing particles were isolated from rice bran by a combination of a differetial centrifugation and an aqueous two phase system, using dextran 500 and polyethylene glycol 6000. The isolated particles consisted mainly of phytic acid, potassium and magnesium. The chemical composition and electron microscopic observation of the isolated particles confirmed that the electron dense material, which is embedded in aleurone particles of matured rice grains, is potassium and magnesium salts of phytic acid.

The proliferating activity of Ehrlich ascites tumor cells decreased with the increase of the concentration of DEPC per 3×10
7 cells and disappeared at more than 200 μg. Sixty per cent of the mice immunized with 200 μg DEPC-treated Ehrlich cells twice survived the challenge inoculation. And employment of Freund's complete adjuvant on the immunization increased the survival ratio up to 90_??_100%.
Respiratory activity, trypan blue stainability and agglutination by concanavalin A were investigated. These results and the electronmicroscopical observation suggest that the specific structure of the cell surface must be maintained in the DEPC-treatment in order to display immunogenicity.
