An iron-oxidizing chemoautotrophic bacterium (
Ferrobacillus ferrooxidans, strain SPP) and a heterotrophic type bacterium (strain G) were isolated from an acid mine water.
Iron-oxidizing activity of whole cells and cell-free extracts of strain SPP was examined from several angles.
Strain G had a strong resemblance to strain SPP, but it hardly grew in inorganic iron medium on transferring directly from organic (glucose) medium.
Though the cell yield of strain G in glucose medium was high (0.8g/
l), the specific activity of iron oxidase of cells was extremely low.
However, it was able to induce iron oxidase in resting cells of strain G harvested from glucose medium by proper treatment with ferrous sulfate.
Therefore, it is possible to obtain abundant bacterial cells which contain high activity of iron oxidase in such way.
