    As regards pharmacologic actions of promin and related preparations on leprosy, the author obtained from experiments made in leprosy partients and animals, following results:
    1) Antibacterial effects of promin, promizole, diasone and methyl-promizole In vitro diasone, promizole and methylpromizole showed nearly equal bacteriostatic action upon tubercle bacilli (Typus humanus H 37 R V), while promin is in its effect a little inferior to them. No difference was seen between promin and diasone in their bacteriocidal action. Promin seemed to be more effective than others on rat leprosy.
    2) Absorption and excretion: When intravenously or orally administered in man, all of these preparations are absorbed and excreted in urine rapidly, although the excretion rate only of diasone is somewhat lower. Promizole is a little more acetylated in human body than others.
    3) Cbncentrations in orga-tissues: The concentrations of each preparation in nerve-tissues are Iower than those in skin. tissues. promin accumulates in organ-tissues somewhat slower than others. Contents of promizole and diasone in various organ-tissues are higher than those of promin
    4) Influences on blood: Though each drug caused anemia, influence of promin was somewhat slighter in comparison with those of others. Resistance of erythrocytes decreases after giving each drug continously for 1 to 2 weeks.
    5) Influences on circulatory organs: No change was seen in blood pressure. The capillary permeability of skin has a tendency to increase after administration of all drugs, while microscopic findings of skin capillaries and skin temperature showed no change.
    6) Influences on reticuloendothelial system: In animal experiment promin activates the function of this system slightly. No definite result was obtained in experiments with other preparations.
    7) Influences on metabolism: No change was seen in the basal metabolism, while protein-, hydrocarbon-, calcium- and cholesterinmetabolism are all disturbed temporarily.
    8) Influences on vitamin metabolism: Decrease of blood level of vitamin B1 was observed after administration of all drugs.
    9) Influences on liver; No remarkable change were seen in Takada's reaction, hepatosuiflein-test and other examinations,
    10) Influences on skin reactions: Q. R, Z. and Mituda's reaction are not influenced.
    11) Influences on acid-base balance: CO2 combining power of blocd decreases sometimes and it leads to acidosis.
    12) Influences in phagocytosis of leucocytes and on electric mobility of erythrocytes The temporary increase of phagocytic funtion of the fomer was seen, while no remarkable change was to be found according to the latter.
    13) Estimated from the results of slide-cell-culture, antibacterial effect of serum on tubercle bacilli is found to be not proportinal to the blood level of each, previously administered preparation.
    14) The serum of patient treated with promin inhibits the Oxygen. consumption of liver of rat.
    15) Diamino-diphenyl-sulfon and its derivates seemed to be generally antagonistic to P A B A-
    16) Diamino-diphenyl-sulfon inhibits the oxygen consumption of emulsion of leproma:

    The following results were obtained in the treatment of leprosy, including 54 1.lepromatous patients, 3 1.maculosa and 1 1.nervosa, with promin and other sulfone drugs since 1947 over the period of four years, investigating the histopathological tissues of skin.
    1) Promin, promisol and diason prevented the bacilli from multiplicating, degenerating and reducing them with vital force.
    2) Simultaneously with the provention of bacilli multiplication, proliferation of the connective tissure and cicatrization of the lesions were observed.
    3) Leprous cells, which had been observed, before the treatment, round, oval and polymorphous with projections, got flatter and spindle-formed and were reduced in number. Thier position against the epidermis changed from perpendicular to parallel in accordance with lesion absorption. And in these cases lipoid corpuscles and vacuole degeneration were seen.
    4) The leprous infiltration extended in the cutis before the treatment were observed atrophic, separated from each other, arround vessels, sweat glands and nerves. But the bacilli were lurking in nerves for a long while.
    5) These processes were not specific only with the treatment of sulfones, but were seen with natural heeling or Chaulmoogra oil treatment, and there may be no essential difference between them.
    6) There are possibility of relaps in future, so further observations, at least 10 years, are necessaly.

    The results of treatments with promin of 714 leprous pationts, including 552 of lepra tuberosa, 112 of 1. nervosa and 50 of 1. maculosa, over the period from 4 months to 4 years were as following :
    1. As for the resorption of nodes or infiltrations, 396 (71.7%) were improved, 1.49(27. 0%) unchanged and 7 (1.3%) aggravated.
    2. Leprouse ulcer : 264 (69.1%) of 382 were healed or improved, 102 (26.7%) unchanged and 16 (4.2%) aggravated:
    3. Leprous macula : 42 (84.0%) of 50 were improved, 7 (14.0%) unchanged and 1 (2.0%) aggravated.
    4. Neuralgia in lepra tuberosa : 70 (15.4%) of 456 were improved, 266 (58.3%) unchanged and 120 (26.3%) aggravated.
    5. Neuralgia in lepra nervomaculosa : 11 (9.8%) of 112 were improved, 75 (97.0%)unchanged and 26 (23.0%) aggravated.
    6. Anaesthesia in lepra tuberosa : 49 (9.2%) of 433 were improved, 302 (56.7%) unchanged and 1.82 (34.1%) aggravated.
    7. Anaesthesia in lepra nervosa : 6 (5.4%) of 112 were improved, 80 (71.4%) unchanged and 26 (23.2%) aggravated.
    8. Anaesthesia in lepra maculosa ; 4 (11.1%) of 36 were improved, 19 (52.8%) unchanged and 13 (36.1%) aggravated.
    9. Motorial paralysis in lepra tuberosa : 54 (10.3%) of 525 were improved, 232 (44.2%) unchanged and 239 (45.5%) aggravated.
    10. Motorial paralysis in lepra nervosa : 10 (9.0%) of 112 were improved, 66 (59.0%) unchanged and 39 (32.0%) aggravated.
    11. The onset of Erythema nodosun was seen in 403 cases (73.1%) of 552 lepra tuberosa treated with promin.
    12. Examination for leprosy bacilli in nasal smear of 104 cases :

    13. Leprosy bacilli can not be easily extinguished, from tissure. In one case they disappeared after a year and eight months.
    14. The toxic manifestations were general weakness, anorexia, anemia and urticaria, and of light degree.
    15. In rare cases the Mitsuda-reaction changed positive, viz in one case it showed moderately positive and in three cases weakly positive.
    16. The results of promizol treatment on 74 cases were favorable and its toxic manifestations were weaker than those of promin.

    We observed the onset of murine leprosy after inoculation anb the corresponding serum fractions with electrophoresis. As the results, the tendency of γ-globulin decrease was seen, instead of the increase of it, which is thought specific in lepra tuberosa.

    The administeration of Tb-1, 30 50mg daily in the beginning of treatment, 200-300mg afterwards and 5-50mg in total to 12 leprous patients inculding lepra nodosa 4, 1.maculosa 6 and 1. nervosa 2 gave us the following results.
    1) Intoxication of the drug was slight anorexia and liver damage, but they did not demand discontinuance of the administration.
    2) The effectiveness was, slight improved in 6 cases, remarkably improved in 2 cases, somewhat effective in 3 cases and indistinct in 3 cases.
    3) Resorption of leprous lesion and transformation of bacilli were recognized clearly from the pathohistological view.
    4) Larger dose of this drug can be recommended in these leprous cases than in tuberculosis.
    5) If, these beneficial effects continue, this drug should be regarded as one of the most promising as the sulfones for the treatment of leprosy.
