    We have been studying glial progenitor cells by using antibodies against the NG2 proteo-glycan and the alpha receptor for platelet-derived growth factor (PDGFa R). In vitro the two mole-cules are expressed on oligodendrocyte progenitor cells but are down-regulated as they differentiate into mature oligodendrocytes. In vivo, they are found not only during development but also exist abundantly throughout the gray and white matter of the mature brain and are distinct from astrocytes or microglia. A small fraction of NG2+ cells in the early postnatal brain differentiates into oligoden-drocytes in vivo. We examined changes in NG2+/PDGFα R+ cells in the dysmyelinating mutant jimpy, which arises as a result of a point mutation in the proteolipid protein gene and is characterized by severe dysmyelination accompanied by premature oligodendrocyte death. In the spinal cords of jimpy mice at postnatal day 18, there was a three- to six-fold increase in the number of NG2+ glial cells that had incorporated bromodeoxyuridine compared with that in the wild type spinal cord. To identify signals responsible for the increased proliferation of oligodendrocyte progenitor cells, mRNAs isolated from jimpy and wild type mice were assayed for various cytokines. There was no significant change in the levels of PDGF A or TGF-β, but there was a significant increase in the level of the che-mokine GROα in the jimpy spinal cord. We are currently testing the possibility that GROα directly acts on glial progenitor cells and stimulates their proliferation.

    To obtain a precise seroepidemiological understanding of measles antibodies, we analysed hemagglutination inhibition (HI) and neutralizing test (NT) antibodies in different age groups in Urawa City. We examined 1591 serum samples collected from subjects aged from zero day to 95 years. Of these 1591 samples, 1094 were HI antibody positive with a positive rate of 68.7%. Mean HI anti-body titer by age was from 240±082 to 254±1.43. Of the 497 samples which were HI antibody negative, 419 were NT positive. HI antibody positive rate and mean antibody titer were decreased at ages 6 to 12 months, and then increased with age. Approximately 5% of school children aged 6 to 15 years were not immune status. The key issue for measles eradication is how to immunize this population.

    In bilateral cleft lip repair numerous complicated problems still remain. In our opinion, it is better to begin treatment soon after birth. We start nonsurgical correction of a nasal deformity using a nose retainer and preoperative orthodontics using a palatal plate. Surgical repair of the lip is done within one month of birth, by which time the nose alveolus and projecting prolabium can be adequately changed into the desired form. Primary lip repair is performed in one stage which includes restoration of the muscle union, labial sulcus reconstruction and nasal correction. We use a two straight parallel suture line method for symmetric bilateral cleft lip repair. The distance between the two cupid's peaks is 4mm. The central tubercle is reconstructed using bilateral cutaneo-mucosal flaps. The tissue volume of both sides is not the same in asymmetric cases so we have refined our method in order to achieve symmetry.

    A comparative study on isolated guinea pig hearts was carried out to determine the role of selenium enriched reperfusion solutions on postischemic reperfusion injury. The hearts of 20 control and 20 study group guinea pigs were mounted on a Langendorff perfusion apparatus and were perfused by gassed Krebs-Henseleit solution at 37°C. The hearts were then arrested by discontinuing the Krebs-Henseleit perfusion. After 20 minutes of normothermic ischemia, in the study group, the hearts were reperfused by selenium enriched Krebs-Henseleit solution (10-3 mMol/L) and in the control group only Krebs-Henseleit solution was used. Postischemic percentage recovery of mechanical cardiac functions (heart rate and contractile force) and postischemic tissue degeneration indicators Malondialdehyde (MDA) and Adenosine deaminase (ADA) were compared between the groups. Mean percentage change of heart rate, contractile force and heart work were significantly higher (p≤0.0001) and tissue MDA and ADA levels were lower (p≤0.001) in the selenium reperfused group. Our results demonstrated that addition of selenium to reperfusion solutions significantly improved cardiac functional recovery and decreased postischemic myocardial injury.
