    Sendai virus infection was examined by the immunofluorescent method in the nasal mucous membrane (NMM) of the mice having received each of five kinds of prophylactic treatment by direct or contact exposure. Administration of interferon or poly I:C via the nasal route decreased in number the viral-antigen-positive cells in NMM in the postexposure period, and the latter treatment protected the lower respiratory organs from contagion. Immunoprophylaxis of the entire respiratory organs against contagion was nearly complete with 6 dosages of UV-inactivated vaccine via the nasal route, but not by administration of alginate adjuvant UV-vaccine via the percutaneous route or ether-inactivated vaccine via the nasal route. However, the adjuvant vaccine protected the lower respiratory organs against contagion. Except for one successful UV-vaccine group, the test-positive cells in NMM of the five vaccinated groups and control increased markedly till postexposure day (PED) 5 and 10 by direct and contact infection, respectively, and thereafter decreased slowly. Viral antigen of the trachea and lungs disappeared on PED 7 and 12 with the development of serum antibody after infection by the two methods. This investigation of viral-antigen-positive cells in the entire respiratory organs, particularly in NMM differentiated in topical immunity and the viral infection between the two regions.

    A total of 241 strains of Pasteurella multocida was isolated from pigs in Japanese field during 1981/82, and was investigated for their dermonecrotic activity for guinea pigs. The strains having the activity were designated as dermonecrotic toxin (DNT) positive (DNT+), and those lacking the activity as DNT negative (DNT-). Of the 241 strains, 62 strains (25.7%) were DNT+ ; 61 were isolated from nasal cavities of a live or dead pigs, and the remaining one was from gross pneumonic lesion. All the DNT+ strains belonged to Carter's serovar D. There were no differences in biologic properties between the DNT+ and DNT- serovar D strains of P. multocida except for dermonecrotic activity. Of all the pigs which showed gross lesions of atrophic rhinitis (AR) and were positive for serovar D strains of P. multocida isolation, 29.5% harbored DNT+ and 26.8% had DNT- strains (P>0.05). Seventy percent of the same population harbored both DNT+ strains of P. multocida and Bordetella bronchiseptica, while 19% yielded only DNT+ strains of P. multocida from nasalcavities (P<0.01). Gross lesions of AR were foundin 12.5% of the vaccinated pigs with commercial AR vaccines, whereas the lesions were found in 92.3% of an unvaccinated pigs. These results suggested that appearence of AR in pigs is due to B. bronchiseptica infection. The DNT+ strains of P. multocida might not be a primary causative agent for swine atrophic rhinitis in Japan.

    Serotypes of 258 isolants of Erysipelothrix rhusiopathiae from slaughter pigs affected with chronic erysipelas during a period from 1980 to 1982 in Japan were determined. Predominant serotypes of 213 isolants from the cases of arthritis or inflamed lesional lymph node were serotypes la, lb, 2, 6, and 11 (12.2%, 7.5%, 71.4%, 3.3%, and 2.3%, respectively). The other serotypes 3, 5, 8, 21, and type N composed 3.3% of the isolants. Of 30 isolants from the cases of endocarditis, 3 belonged to serotype la, 2 belonged to serotype 1b and 25 belonged to serotype 2. Of 15 isolants from the cases of urticaria, 14 belonged to serotype 2 and I to serotype 5.

    Regularinterferon (IFN) was detected in the sera of mice infected with Toxoplasma gondii, RH or S-273 strain, only 24 hr postinoculation (p.i.). Mice which survived from subchronic (14 days p.i.) or chronic (more than 40 days p.i.) toxoplasmosis, were injected intraperitoneally with Toxoplasma lysate antigen (TLA), and IFN-γ was detected in their serum 6 hr after injection. No serum containing regular IFN inhibited Toxoplasma multiplication in mouse macrophage monolayers in vitro. The molecular weight (m.w.) of regular IFN and IFN-γ in the serum was mainly between 40, 000 and 70, 000. The highest regular IFN activity produced by normal spleen cells cultured in vitro was contained in fractions with approximate m.w. of 40, 000. It was contained in the portion of alpha-globulins of the serum. The IFN activity in L-929 cells cultured with poly I:C was contained in fractions with m.w. of 24, 000 to 70, 000. The highest IFN activity was observed in samples with approximate m.w. of 60, 000. IFN-γ contained in spleen lymphokines was estimated to have approximate m.w. of 55, 000 to 70, 000, with maximum activity with a m.w. of 67, 000.

    The effects of the intravenous injection of arginine (1.25 mmoles/kg) on plasma insulin, glucagon, 11-hydroxycorticosteroids (11-OHCS) and glucose were compared in castrated Landrace piglets between the thermoneutral (TN, 22°C) and the cold environment (2°C) groups with constant relative humidity (60%). Plasma insulin rose transiently from 8 μg/ml in initial level to 17 μU/ml 5 minutes after the arginine injection in the TN environment. Likewise, plasma glucagon rose from 122 pg/ml to 253 pg/ml. The cold exposure augmented the insulin and the glucagon responses to arginine. The 11-OHCS level before the arginine injection was 6.5 μg/dl in the TN environment. It increased significantly 4 and 7 days after cold exposure (11.9 μg/dl and 9.4 μg/dl). The arginine injection in the TN environment induced plasma 11-OHCS and glucose. These responses to arginine were augmented by the cold exposurd. The potentiation of arginine-induced insulin secretion by the cold exposure in piglets is contrast to the previous resultes obtained in sheep.

    Haemophilus pleuropneumoniae strains isolated from pigs during the period from 1974 to 1982 were serotyped and examined for their susceptibility to 21 antimicrobial drugs. Of 109 field isolates in Japan and surrounding countries (Japan; 105, Korea; 2, Taiwan; 2), 105 strains were of serovar 2, two were of serovar 3, and the remaining two were of serovar 5. The great majority of the field isolates, 107 strains, were highly susceptible to penicillins, chloramphenicol, tetracyclines, rifampicin, trimethoprim and furazolidon. These strains were also susceptible to cephalexin and colistin. However, susceptibilities to aminoglycosides, macrolides, sulfamonomethoxine and nalidixic acid were not so high. The ramaining two strains were found to be drug-resistant; strain Hp1 was resistant to penicillin, tetracycline, streptomycin and sulfonamide, and strain 966 was resistant to streptomycin and sulfonamide. The former was identified as serovar 3 and the latter was serovar 5. All the 105 strains of serovar 2 were durg-sensitive, and no increase in the resistance of this organism had been observed during the period from 1974 to 1982.

    Of 21 drug sensitive Escherichia coli strains isolated from laboratory mice and rats, 17 harbored 1 to 9 cryptic plasmids. To examine the convertibility of these cryptic plasmids into conjugative R plasmids, we introduced a nonconjugative plasmid, pMK1::Tn2602 (ColE1::Tn5::Tn2602), which encodes resistance to kanamycin (Km) and ampicillin (Ap), into a mouse strain E916 possessing at least 5 plasmids by transformation. A mixed culture of the transformant E 916 (pMK1::Tn2602) as donor and E. coli K-12 strain as the recipient produced Km-resistant and Ap-resistant transconjugants at the frequencies of 10-9and 10-6 per donor cell, respectively. Agarose gel electrophoretic analysis revealed that the Km-resistant transconjugant had a single plasmid (pKO916Km) of 29.5 megadaltons (Mdal), which was capable of transforming another strain of E. coli K-12 into a Km-resistant. Similarly, the Ap-resistant transconjugant had a single conjugative R plasmid (pK0916Ap) of 29 Mdal. Restriction enzyme cleavage and electron microscopic analysis demonstrated that these R plasmids were the composite plasmids consisting of a 26 Mdal cryptic plasmid presented in the strain E916 and either of a Km transposon (Tn5) or an Ap transposon (Tn2602). They were conjugally transmissible to different species of enterobacteria such as Shigella and Salmonella. In vivo transfer of pKO916Ap was also demonstrated in gnotobiotic mice.

    Morphology and distribution of the juxtaglomerular (JG) cells of chicken kidneys were ontogenetically studied by light and electron microscopy. JG cells contained fine secretory granules having a diameter of about 350 nm and myofibrils in their cytoplasm. Following three types of JG cells were classified tentatively by their positions for the convenience of description: 1) A type: located at the walls of the glomerular arterioles; 2) V type: located at the paravascular poles; 3) M type: located in the mesangial regions. There were no morphological differences among the granular characteristics of the three types of JG cells; however, it was noted that some JG cells of M type showed obscure association with neighboring basement membranes of glomerular capillaries and contained almost no myofibrils in their cytoplasm. Ontogenetically, JG cells were initially detected in embryos of 19 days of incubation, and their histoplanimetrical frequency was increased until the first day of posthatching life. However, a prominent decrease in the frequency was characteristically estimated at the second posthatching day. Thereafter, their relative histoplanimetrical values were observed to tend to increase constantly. Concerning the frequencies estimated among the three types of JG cells for the period of 1 to 15 posthatching weeks, the relative frequency of the glomeruli containing A type of JG cells was clearly decreased in contrast with that of M type. However, there was no conspicuous changes of relative histoplanimetrical values in the glomeruli containing V type of JG cells throughtout the investigated period.

    In Babesia rodhaini infection in rats parasitemia remained up to 3 weeks after clinical recovery. The immune responses were demonstrated at least up to one year after recovery.

    The abdominal organs and the fetus of normal cows, horses, goats and dogs were investigated by the use of ultrasonotomography. 1) Liver & gall bladder: In cows and goats, the liver was imaged on the right side between the 8th and 12th intercostal spaces. Clear ultrasound images of the canine liver were obtained by subcostal scanning. In horses, however, only the parenchymal margin was visualized. In cows, goats and dogs, the parenchyma, portal vein, hepatic vein and gall bladder showed characteristic echo patterns, respectively, but the appearance of the bile ducts was obscure. 2) Spleen: In each animal, the ultrasound images of the spleen were obtained on the left side in the lower intercostal spaces; in horses and dogs, the same was obtained also in the flank. The spleen appeared as the characteristic low amplitude echo pattern. 3) Kidney: In cows and goats, each kidney was displayed on the right flank, and the right kidney was also observed in the 12th intercostal space. In horses and dogs, each kidney was visualized in the lower intercostal spaces and the flank. In all animals, echoes of the kidney were composed of a low amplitude echo pattern showing the renal cortex, an echo free pattern exhibiting the medulla and an echogenic pattern indicating the pelvis. 4) Fetus: The fetus and fetal organs of cows, goats and dogs were imaged by transcutaneous scanning in the lower abdomen or flank. Fetal movement and fetal heart contractions were detected by real time ultrasongtomography.

    During anaerobic fermentation of the filtrate juice of swine feces, mastigophoran and sarcodine protozoa were observed, while no ciliate protozoa appeared. Monocercomonas spp. and Amphimonas spp. of mastigophoran protozoa as well as small amoebae of sarcodine protozoa were predominant.

    Oral administration of pyrimethamine during the second half but not during the first half of the organogenetic period caused a high incidence of the cleft palate in rats. The findings being different from those in pigs suggest that the critical period might be species specific.

    The distribution of hydroxysteroid dehydrogenase (HSD) was histochemically examined in the labyrinth of the dog placentae. Except at term, weak activities of Δ5-3β-HSD were detected in the trophoblast without change in activity during pregnancy. By electron microscopy neither cytotrophoblast nor syncytiotropholast seemed to be a steroid-secreting cell. The results suggested that the trophoblast cannot synthesize steroid hormones de novo in the dog placenta.

    Localization of nerve ending in the corpus vasculare paracloacale (CVP) of the cock was studied electron microscopically. Two types of nerve ending were observed in the CVP. The first type contained small clear vesicles and large dense cored vesicles about 50 and 100 nm in diameter, respectively, while the second type contained small dense cored vesicles about 50 nm in diameter and large dense cored vesicles. The former was observed adjacent to pericytes or capillary endothelial cells and the latter was adjacent to small arteries.

    An astrocytoma occurring at the putamen/nucleus caudatus, thalamus and mid brain was experienced in a male 24-month-old Wistar rat. Ultrastructually, tumor cells showed abundant subcellular organelles with lipid droplets in the cytoplasm and amount of glial filaments in processes.

    Total and ionic calcium concentrations in maternal and fetal blood and amniotic fluid from normal rat pregnancy were determined. Total calcium concentrations in maternal and fetal blood fluctuated during gestation, while the ionic calcium concentrations were of almost constant level, being 1.19∼1.26 mmol/l in maternal blood and 1.60∼1.67 mmo1/1 in fetal one. The role of non-ionic calcium and amniotic calcium in relation to the constant ionic calcium level was discussed.

    Isolation and serological survey of Chlamydia psittaci was performed in feral pigeons and crows captured in Hokkaido. Complement fixing antibodies were found in 32(30.5%) out of 105 pigeons, and in 18(22%) out of 82 crows. The organisms were isolated from the spleens of 9(7.6%) out of 119 pigeons but not from 179 crows.

    Amyloidosis was observed in 3 out of 6 male rabbits experimentally infected with Trypanosoma evansi. Amyloid deposit was seen in the splenic perifollicular spaces, hepatic sinusoids or renal glomerular tufts. Lymphoid and plasma cell clusters were seen in the spleen and lymph nodes with persistent high levels of serum IgM.
