    Attempts were made to determine the position of the portion on the structure gene for amylase specifying the electrophoretic characteristics of amylase-4, amylase-1088 and amylase-1 produced by different strains of Bacillus subtilis. The results showed that each of amy4, amy1088 and amy1, which were allelic to each other, might have respectively a portion, namely E4, E1088 and E1, locating at a definite position between O5 and A4, and specifying the electrophoretic characteristics of each amylase protein. Data were obtained which showed that E might occupy a certain length on the structure gene.

    The karyotypes of Trillium tschonoskii (2n=20, K2K2TT) collected from four localities of Japan Islands have been studied by means of differential reactivity of H-segments of the chromosomes to low temperature.
    In all 14 plants examined, 10 pairs of chromosomes were found to be homozygous. But the karyotype of plants from Mt. Aso (Kyushu) was different with regard to chromosome D from that of the plants from the other three localities, Hayakita and Ashoro in Hokkaido and Mt. Tsurugi in Shikoku.
    The above findings suggested that one of the parental, now extinct, diploid species (2n=10, K2K2) might be differentiated into homozygous local populations. The other extinct homozygous diploid species (2n=10, TT) had hybridized with K2K2 at different localities to give rise to the present T. tschonoskii.

    Chromosome constitutions of callus cells originating from the root of tobacco and from the root and the ovary of wheat were studied. Effects of duration of subculturing and of exogenous substances added to the medium on the chromosome number of callus cells were also studied. In tobacco, the chromosome number in callus cells derived from root tips, subcultured for three to seven months, was predominantly diploid, while in the two year-subcultured callus chromosome numbers varied widely. In common wheat the chromosome constitution of callus cells derived from root tips subcultured for two to four years was similar to that of callus cells from the ovary; in both diploid cells were predominant. The chromosome constitution of callus cells which originated from the root tips of emmer wheat and were maintained on media supplemented with casein hydrolysate was quite similar to those maintained on media containing kinetin, adenine sulfate and 2, 4-D.

    Pollen grains with Bz-gene of maize were exposed to gamma-rays in single or two equal fractions and crossed to recessive stocks to observe mutations to bz F1 seeds. A linear dose-mutation frequency relationship was confirmed up to 3000R in the sum of whole and partial mutations expressed as bronzy aleuron color. While the frequency of partial mutations did not show any significant difference by single or fractionation treatment, that of whole mutations was significantly decreased by fractionation with an interval period of 120 minutes or less. No temperature effect was found by holding at 5-30°C. It is concluded that the mutagenic action of gamma-rays was reduced by dose-fractionation probably due to certain recovery events taking place under experimental conditions applied.

    The pattern of development of amylase activity during growth of a transformable B. subtilis 168 grown at 52°C was different from that observed at 37°C. Amylase was produced during the stationary phase, but not at the exponential growth phase at 37°C, while, at 52°C, it was produced during the exponential growth phase, but not at the stationary phase. A type of mutants, whose pattern of development of amylase activity at 37°C was quite similar to that observed when B. subtilis 168 was grown at 52°C, were isolated from B. subtilis 168. They exhibited alterations of different phenotypic traits, such as loss of spore forming ability and altered pattern of cell growth, which were also found when B. subtilis 168 was grown at 52°C. Genetic studies showed that these phenotypic traits might be controlled by a single gene. The fact that one of these mutants was found to have a defect in the citric acid cycle, suggested an important role of functional citric acid cycle on the regulation of amylase production, especially in specifying the pattern of development of amylase activity during growth of B. subtilis 168.

  • III. HOMOEOLOGY BETWEEN GENOMES A (x=8) AND B (x=9)

    The relationship between the genomes A (x=8) and B (x=9) of Scilla scilloides complex has been studied in F1 hybrids (AB, 2n=17) between the two diploid types AA (2n=16) and BB (2n=18).
    Pairing of chromosomes is variable among PMCs, the maximum number of bivalents per cell being 8. Some bivalents are clearly heteromorphic. The pairing of chromosomes in PMCs revealed that the genomes A and B are segmentally homologous, although they are karyotypically well differentiated.
    A scheme is presented to interpret how the genome A has been differentiated from the genome B by translocation, inversions, and loss of a centromere.
