In our field, antifungal antibiotics chiefly Trichomycin, Nystatin, Amphotericin B have been used and we have reported the results of clinical experiments of those drugs, but we also carried out its preventive experiments and its in vitro antifungal experiments to prove its usefulness in clinical treatment. On the other hand, we studied inhibitory effect of respiration on
Candida by those antifungal antibiotics and we found it is pararell with other effects to fungus.
Most of the blood level tests of those antifungal drugs have been reported as negative due to inactivation effect of blood elements on those drugs. But we succeeded to observe it on Trichomycin by using micronephelometer.
We studied the antifungal effects of those drugs on the changes of morphology and inner structure of fungus by using phase-contrast microscope, and we observed the atrophy of fungus by touching with those drugs and without destruction. This phenomena was particularly prominent with Trichomycin and Nystatin. Irregularity of internal membrane and mitochondria were observed as the changes of inner structure.
To observe the influence of those drugs on the metabolism of fungus, Trichomycin was used on
Candida albicans. It was presumable that its inhibition was under glycolytic system because Trichomycin inhibited all oxidation of saccharide. Also its inhibitory point was presumably in TCA-cycle since it has strong inhibitory effect on organic acids on TCA-cycle or on the aminoacids which can be directly converted to TCA-cycle.
Although we cannot be sure about the turnover inhibition of ATP only by the presence of incorporation inhibition to the acid soluble phosphate fraction of P
32, we cannot deny the inhibition of production of ATP.
We are sure now that Trichomycin is working on the mechanism of protein synthesis because it showed marked incorporation inhibition on nucleic acid fraction and also due to the fact that nucleic acid concerns to the protein synthesis and our observation of incorporation inhibition of S
35-methionine to the fungus.
Regarding the resistance for antifungal antibiotics, it has not been our problem yet, but we cannot be negligible in future about its increasing tendency of the widely used antifungal antibiotics.
