    A simplified slide preparation for the circumoval precipitin (COP) test for field use is described. The test makes use of an insulating tape and cellulose tape in place of coverslips and also makes use of Schistosoma eggs dried on slides. S. japonicum was used in this study, and dried eggs gave satisfactory COP reaction up to 2 month strorage. Use of tapes was found to be more suitable for field use and, at the same time, cheaper than coverslips.

    Cultured rabbit alveolar macrophages release various fatty acids, prostaglandins and other arachidonic acid metabolites upon stimulation. The selectivity of fatty acid release depends on the stimulus used. Zymosan, a phagocytic inflammatory stimulus, causes a fourfold increase in prostaglandin release, and a threefold specific release of arachidonic acid. Phorbol myristate acetate, causes a fourfold increase in prostaglandin release, and a non-specific release of long-chain fatty acids. Bacterial lipopolysaccharide leads to non-specific release of the long-chain fatty acids, but a minor increase in prostaglandin production. These data suggest that fatty acid release during phagocytosis is a specific cellular response rather than non-specific lipid hydrolysis. The potential significance of fatty acid release in macrophage function was discussed.

    A simple procedure for obtainig drug-resistant strains of Acholeplasma, occuring naturally and spontaneously at a low frequency in a broth culture was described. The drug-resistant strains could be obtained and enriched for by the addition of more than twice volumes of fresh broth containing antibiotics to a pre-cultured medium with Acholeplasma at the plateau phase of the growth. Furthermore, the growth of drug-resistant cells was promoted by the addition of broth containing protease and an antibiotics. By means of this procedure, various kinds of antibiotics-resistant strains of Acholeplasma were easily isolated, some of which were not obtained by the agar plate selection method. The merits of this procedure were that the scale of this system could be controlled and that the isolation of drug-resistant strain was very simple and rapid.

    A bovine enterovirus (MZ-468) was adsorbed to Sarcoma-180 cells at 4°C. The cells were reacted with FITC-labeled IgG antibody for the virus and were studied with a laser fluorescence cytoflowmeter. From the histograms, binding-saturation of receptors was observed and the dissociation constant (Kd) was estimated. A new binding rate constant was deduced from the change of the ratio of virus-adsorbed cells against time and virus concentration. The constant has a dimensions of (cm3/min·receptor-site) and probably represents the receptor class on a cell with the highest rate constant.

    The properties of a toxin produced by Propionibacterium acnes isolated from patients with Kawasaki disease were investigated. The toxin activity declined gradually during repeated passages in GAM broth but could be recovered by resuspending with fresh GAM broth after ultraf iltration. A larger increase of toxin activity was obtained with digested serum powder than with GAM broth. Inactivation experiments suggested the presence of two components with toxin activity in the culture filtrate. Analysis of the culture filtrate by polyacrylamide gel electrophoresis also demonstrated the presence of two proteins which might be responsible for the toxin activity.

    The authors have designed the crossed, double figure-of-eight wiring (CD-8 wiring), which was compared physically with other types of wiring, and it was confirmed that the method designed was the strongest. Operative procedure is as simple as the ordinary figure-of-eight wiring and a firm fixation of the fragments is secured. The authors recommend CD-8 wiring for treatment of fractures, for which figure-of-eight wiring would originally be indicated.

    A case of congenital deficiency of factor XIII (fibrin-stabilizing factor) with traumatic hematoma of the posterior fossa which was successfully treated is described. Congenital deficiency of factor XIII is a quite rare coagulation disorder and 100 or more cases with this disease have been reported. We have found a few cases undergoing craniotomy for associated intracranial hematomas. In this report, the diagnostic and therapeutic problems associated with this rare coagulation disorder are also briefly discussd.

    Peripheral blood mononuclear cells from 45 children, aged 1 to 19 years were evaluated for natural killer cell cytotoxic activity against K-562 erythroleukemic cells, The results of a 51Cr release assay demonstrated that the NK cell activity of normal children is dependent on age and sex. Younger females, aged 1 to 9 years, were more reactive than elder females aged be-tween 10 and 19 years (P<0.001). Males aged 1 to 19 years were less reactive than adults (0.02<P<0.05). Among children, the levels of NK cell activity were significantly greater in females than in males (0.001<P<0.01). In contrast, among elder children, the levels of NK cell activity were significantly greater in males than in females (0.01<P<0.02). The NK cell activity of elder female children was 60% and 59% that of the elder male children, at the two E:T ratios of 50:1 and 100:1, respectively.

    Menetrier's disease is a relatively rare disease characterized by greatly enlarged gastric rugae resembling the convolutions of the brain and often associated with hypoproteinemia. Although radiographic examination of the stomach is one of the most reliable diagnostic measures of this disease, it is hard to clarify the structure of the gastric wall by this method. Ultrasonographic examination of the stomach, which has recently become more practical, has the advantage of delineating the structure of the gastric wall. We report here the ultrasonogram in a case of Menetrier's disease. The ultrasound demonstrated gastric wall thickening (pseudo-kidney sign), crinkled mucosal folds projecting into the lumen, a smooth serosal surface with a clear boundary between the adjacent organs, and a pliable gastric wall. The follow-up study revealed no changes in the sonographic appearance. The diagnostic value of ultrasound in Menetrier's disease and its limitations are discussed.

    The simultaneous determination of the biogenic amines, dopamine and serotonin, and their metabolites, 3, 4-dihydroxyphenylacetic acid, homovanillic acid, 3-methoxytyramine and 5-hydroxyindole-3-acetic acid, in brain tissue was investigated using high performance liquid chromatography with electrochemical detection. A clearly separated and highly sensitive chromatogram was obtained. A favorable condition to prevent the reduction of the amines and their metabolites in either the standard or tissue homogenate was the use of 0.1 M formic acid for the stock and homogenizing solutions. The dopamine level in the nucleus caudatus putamen was increased 20 min after treatment with methamphetamine (4 mg/kg, i, p.), but not in other areas. The 3, 4-dihydroxyphenylacetic acid level was reduced in the frontal cortex, nucleus accumbens, tuberculum olfactrium and nucleus caudatus putamen, and the homovanillic acid concentration was decreased in only the nucleus accumbens following methamphetamine. The serotonin and 5-hydroxyindole-3-acetic acid levels were not changed significantly in any of the brain regions. The present observations suggest that methamphetamine, at an early stage after administration, leads to an inhibition of the degradation of dopamine (but not Serotonin) in central nervous system.
