    The plaster containing [14C] methyl salicylate was applied to the back of hairless mice of HRS/J (hr) strain. [14C] Methyl salicylate was percutaneously absorbed and distributed with a high concentration in the skin at the applied site. The peak level (1.5 mg%) of salicylate in the serum was attained 2 hours following application. Approximately 40% of the dose applied was excreted in the urine of 48-hour treated mice. Ten sheets of the plaster containing non-radioactive methyl salicylate (35.0 mg/sheet) were applied to the back of male human subjects for 12 hours. The amount absorbed percutaneously could be estimated to be approximately 37% of the dose applied. The serum concentrations of total salicylates and free salicylic acid reached a peak at 8-12 hours. Within 12 hours after removal of the plaster, practically all of the total salicylates and free salicylic acid were excreted from the body. Application of the plaster for 12 hours a day was repeated for consecutive six days. Trace or no amout of total salicylates and free salicylic acid was detected in the serum 12 and 30 hours after removal of the last plasters. No significant changes were noted in the hepatic functions of all the subjects before and after the repeated application.

  • 3. DETECTION OF CIRCULATING ANTIGEN BY COUNTER IMMUNOELECTROPHORESIS

    Using counter immunoelectrophoresis, an antigen in circulation of rabbits infected with Schistosoma japonicum was investigated. The sensitivity of counter immunoelectrophoresis was found to be 16 to 32 fold higher than that of ouchterlony test. The rapidity and simplicity seemed to be much superior to other serological tests. The antigen was detected in 22 of 24 infected rabbits with various periods of infection. The worm burden in negative cases were very low. In all infected rabbits, the antigen was constantly found since the appearance. The antigen titer in serum of each infected rabbits was significantly correlated with the worm burdens per body weight (r=0.70, p<0.01).

    1. Craniocerebral reference points and the width and height of the third ventricle were measured in intraoparative positive pressure pneumovetriculograms on 71 patients who underwent stereotaxic surgery for various reasons. 2. When reference points were located on two-dimensional plane in relation to the anterior comissure, distant reference points were more variable than those close to the anterior comissure. 3. The CA-CP distance and the width of the third ventricle were 22.9±1.9 and 8.1±2.3 mm (mean±S. D.) respectively and the mean values of the width and height of the third ventricle and the CA - CP distance were greater in males than in females respectivery. 4. A correlation coefficient was high between the width of the third ventricle and the CA-CP distance, but was low between the width and height of the third ventricle and between the height of the ventricle and the CA-CP distance.

    Sympathetic ganglion cell membranes of cold- and warm-blooded animals are depolarized and initiate spontaneous repetitive firing of action potentials in Ca-free solutions containing a chelating agent. The depolarization of cell membranes and duration of these action potentials were increased progressively with time after preparations were perfused with this solution. These cells were able to produce action potentials forming a prolonged plateau phase even after spontaneous firing ceased completely while the membrane was strongly depolarized to -10 to -20 mV, provided membranes were previously hyperpolarized by extrinsic anodal currents.

    An electrophoretic method for determining hyppoxanthine-guanine and adenine phosphoribosyltransferase in a mammalian tissue is presented. The method based on Kizaki's micro-assay procedure was improved with modification using a technic of thin-layer radio-chromatographic scanning. The modification serves to reduce the time in the enzyme assay without any interference among nucleotide, nucleoside and other decomposition product. In this paper, the assay was applied to crude extract of cow snout epidermis, and the salvage enzyme activity in the epidermis was determined.

    Sixteen non-fusing mutants (γ) were isolated from virus (G) possessing fusion ability. Some mutant pairs complemented each other which resulted in an extensive cell fusion as original G virus, while the other mutant pairs did not. Thus, these mutants were classified into four complementation groups. By recombination analysis using eight γ mutants belonging to each complementation group, a linkage map has been constructed with regard to cell fusion. It has been demonstrated that four or possibly six cistrons are associated with the fusion event.

    Day-to-day observations were made on the ureterograms after extensive hysterectomy in cancer of the cervix to reach the conclusion as follows. (1) Combined use of X-ray television is advantageous in detecting ureteral impairment after operation on cancer of the cervix. (2) Ureteral impairment after operation on cancer of the cervix starts developing relatively early after operation. (3) It is 3 to 4 days after operation that the impairment is most severe. (4) The impairment heads for recovery after the seventh day, but it persists in serious cases. (5) Judging from the findings above, there is a possibility that the ureteral impairment after operation on cancer of the cervix can be recovered to some extent. (6) Our impression is that if apreventive step for the ureteral impairment is necessary, carrying it out within seven days will be effective.

    There can often be seen stacks of the annulate lamellae in the principal cells of the epididymal epithelium of the Japanese monkey, which are associated with the rough-surfaced endoplasmic reticulum (rER). The fine structure and formation of these annulate lamellae were discussed, and in addition the arrangement of the pore-complexes in this peculiar structure were examined to compare with those of the nuclear pores.

    We carried out a role of horny layer of skin as for skin contamination. In the case of the skin with horny layer, the internal permeance of 131I-Na was slow and slight. In the case of the skin without horny layer, we had better think that the internal permeance of 131I-Na was rapidly and revealed the internal contamination as almost same as in the case of venous injection of 131I-Na. For the prevention of the internal contamination, it was necessary that horny layer of skin was intact. In the case of the normal skin with horny layer, the permeance of 131I-Na was slow by 20 min. after application and then the contamination of 131I-Na should be removed within 20 min. after application. Out of the radioactive substances, iodine, anion, has the easy permeability and volatility. So we have pay expressly attention to handling it comparing with the other radioactive substances.

    A study has been made of the glycosaminoglycans and glycopeptide in the abdominal walls of the rats. The abdominal walls were separated into three layers ; dermis, adipose tissue and muscle containing fascia, and then uronic acid, sulfate, hexose and sialic acid contents were measured in each layer. Values per g of wet tissue were highest in the skin, whereas those of dry tissue were highest in the adipose tissue. Cellulose acetate strip-electrophoresis of materials in barbital buffer revealed the presence of three alcian blue positive fractions in every layer. The relative proportions of the fractions which corresponded in mobility to hyaluronic acid, to heparan sulfate and to chondroitin sulfates were approximately 40 %, 10 % and 50 %, respectively.

    Semisolid culture medium which is composed of inorganic salts only, for the growth and differentiation of Pseudomonas aeruginosa from other Pseudomonas was prepared. When the strains of Pseudomonas aeruginosa tested here were inoculated to the medium, decolorization of medium due to oxidative fermentation took place according to the degree of biological activities. These findings are quite similar to those of PYP medium which is widely commercial as fermentation test of Pseudomonas aeruginosa. From the results, there is a possibility to be used as the medium for fundamental research of metabolism of Pseudomonas aeruginosa, because the medium is composed of inorganic salts, without enriched nutrients.
