  • I. The Changes of DNA, RNA and Protein Contents of Liver After CCl4 Administration to Male Mice

    Changes of DNA, RNA and protein contents of the liver which are the fundamental macromolecules of cells were determined using CCl4-treated male mice, in order to study the molecular mechanisms of liver cell necrosis. The total DNA content of the liver reached a minimum value at 24 hours after the CCl4 administration, and then gradually increased till 72 hours when the value was higher than that at 0 hour. It was postulated that these changes of the total DNA content depended upon either decrease or increase of cells induced by necrosis or regeneration, respectively. The RNA content of the liver per mg DNA also reached a minimum value at 24 hours after the treatment and then increased till 72 hours when the value, however, was lower than that at 0 hour. On the other hand, the protein content of the liver per mg DNA did not varied for the first 24 hours, and then progressively decreased till 72 hours. It was discussed that the regulation of protein biosynthesis of the liver was severely disturbed by the CCl4 administration, and that this disturbance was one of direct factors that caused liver cell necrosis.

  • II. The Decrease of Ribosomes and the Disaggregation of Polysomes of Liver After CCl4 Administration to Male Mice

    It is known that RNA content of liver is greatly altered by CCl4 administration to animals. Therefore, since more than 70% of total RNA of liver are ribosomal RNA, changes of ribosomes of mouse liver by the CCl4-treatment were examined in the present study. The content of membrane-bound ribosomes at 24 hours after the treatment, 0.37+0.02mg/g liver was significantly lower than that of normal liver, 1.10+0.07mg/g liver. However, no significant decrease of free ribosomes occured. A marked disaggregation of polysomes was observed by sucrose density gradient centrifugation after the CCl4 administration. However, no substantial difference of profiles of ribosomal RNA was shown between normal and injured liver. Not only the decrease of ribosomes but also the disaggregation of polysomes gave an evidence suggesting that a serious disturbance at the translational level of protein biosynthesis was induced by the CCl4 administration.

  • III. The Decreased Activity of Factors Working on Translational Level of Protein Biosynthesis of Liver Cells by CCl4 Administration to Male Mice

    It has been reported by several authors that CCl4-treatment of animals caused a decrease of ribosomes and a disaggregation of polysomes of the liver. In the present report, effect on the factors working on the translational level of protein biosynthesis of mouse liver by CCl4- treatment was studied in a cell-free system. A decreased activity of ribosomes of the injured liver was observed in an incubation system without poly (U). On the contrary, no decreased activity was shown by ribosomes of the injured liver when the ribosomes were incubated under higher Mg++ concentration (10mM) using poly (U) as a template. It was postulated, therefore, that the apparent decrease of ribosomal activity of the injured liver was due to an inhibition of initiation factors. Decreased activities of pH 5 enzymes and pH 5 supernatant fractions of the injured liver were also detected. It was discussed that disturbance of the translational level of protein biosynthesis was an essential step to cause liver cell necrosis.

    Leucocyte migration inhibition factor (LMF) against insoluble liver cell membrane fraction (ILM) prepared from rat livers was noted more often in patients with chronic hepatitis and cirrhosis than control persons, (P<0.05) and LMF against rat renal cell membranes was not found in the same patients and control. The patients with LMF to ILM tended to reveal positive cellular response to HBAg in comparison with those without the LMF to ILM (P<0.10). A presence of LMF against ILM, however, seemed not to be parellel to that of circulating autoantibodies.

    Evidence has been accumulated that leucocyte migration tended to be inhibited with rat or human liver mitochondria in some of autoimmune diseases. The inhibition in a presence of outer membranes of rat liver mitochondria was observed in 38% of cases of chronic inactive hepatitis, 39% of chronic active hepatitis and 40% of liver cirrhosis, and in a presence of inner membranes of the mitochondria in 53%, 44% and 36% in the same order. There was no clear parallelism between in a presence of circulating mitochondrial antibody and in that of cellular hypersensitivity against either of mitochondrial membranes. The presence of the later may represent a marker of autoimmune state of chronic liver diseases.

    1) 35S-sulfate was incorporated into the purified collagen extracted from fibrotic rat livers.
    2) Human liver tissue obtained by needle biopsy was incubated with 35S-sulfate. Electron microscopic autoradiography revealed that the location of the particles of 35S-sulfate were mostly coincidental with the presence of collagen fibers.
    3) In fibrotic liver obtained by needle biopsy, regularly aligned ruthenium red positive granules were found on both sides of collagen fibers at an interval of 60 to 70nm.
    4) After partial hepatectomy of fibrotic rats, activities of N-acetyl-β-glucosaminidase and β-glucuronidase in liver tissue were elevated, especially in CCl4 treated rats.
    5) Simultaneous action of collagenase and mucopolysaccharide degrading enzymes during resorption of collagen was suggested.

    The sera of 150 cases of various hepatic diseases and 2350 normal subjects were tested by thyroglobulin and microsome-coated tanned red blood cells hemagglutination.
    Antinuclear factors were tested by the immunofluolescence method and HB antigen were tested by SRID method.
    Thyroid antibodies were present in 2 (33.3%) of 6 cases of lupoid hepatitis, and in 3 (7.7%) of 39 cases of acute hepatitis, and in 2 (7.7%) of 26 cases of active chronic hepatitis, and in 1 (4%) of 25 cases of inactive chronic hepatitis, and in 2 (4.1%) of 49 cases of liver cirrhosis.
    Fifty-one (2.3%) of 2350 cases of normal subjects showed a positive reaction for thyroglobulin antibodies, and fifty-nine (2.5%) showed a positive reaction for microsomal antibodies.
    There was clear relationship between thyroid antibodies and antinuclear factors in various hepatic diseases.
    There was no mutual exclusion and no relationship between HB antigen and antinuclear factors and thyroid antibodies.

    In the present paper 38 autopsied cases with hepatocellular carcinoma were studied, in which bone metastases were verified in 6 cases at autopsy.
    From the symptomatic points of view, all these patients with bone metastases complained initially of bone destruction such as pain in the extremities and/or difficulty in walking. However, from the results of laboratory examination any of data, blood examination and the levels of serum alkaline phosphatase and calcium did not suggest the evidence of bone metastases.
    Microscopical examination of liver showed that 86.8% of heapocellulart carcinomas were associated with cirrhosis and 69.7% of these cirrhosis were postnecrotic type, and that in almost cases histological pattern of hepatocellular carcinoma was classified into Edmondson grade II. However, there was no particular histological pattern which seemed to be dominant in the cases with bone metastases of hepatocellular carcinoma.

    A case of Zieve's syndrome is described in this report with an acute onset of jaundice, hepatomegaly, hyperlipemia and hemolytic anemia based on an alcoholic liver cirrhosis.
    During the course of this case serum alpha-fetoprotein was detected by the micro-Ouchterlony method transiently but HB-Ag was always negative. Although the pathogenesis of this phenomenon is not clear this detected alpha-fetoprotein might be associated with the regenerated hepatocytes in an early stage when the necrotic foci coexist.
    The liver biopsy specimen was characterized by irregular liver cord arrangements and segmentally increased liver fibrosis with slight fatty metamorphosis and partly lobular disorganization, but electron-microscopically were there no enlarged and degenerated mitochondria and no abnormal proliferation of smooth surfaced endoplasmic reticulum.
