Med., 1987, 153 (1), 5-9 - A simple spectrophotometric method for the quantitating of the clot promoting activity of thromboplastin in commercial preparations was developed. Thromboplastin activities were proportional to the maximum rate of clot formation. This was calculated from curves of absorption change during the coagulation of plasma coagulation control. Tohoku J. exp. Med., 1987, 153 (1), 11-20 -Two cases of multiple myeloma which showed inclusions within the cytoplasma of myeloma cells were reported. One contained crystalline inclusions which were not stained by May-Grünwald and Giemsa, and the other contained Auer rod-like spindle shaped inclusions which were stained by May-Grünwald and Giemsa and PAS. Both cases were stained intensively by acid phosphatase but they were not stained by Congo red or by thioflavine-T. Moreover, they reacted only with anti-x sera in the immunofluorescent study and showed the same structure on electron microscopic observation. Although they showed different staining behaviors according to their compositions, they were considered to be essentially the same. It appears that light chains produced in excess are concentrated and form inclusions through the addition of sugar or digestion by lysosomal enzymes.抄録全体を表示PDF形式でダウンロード (6958K) -

Localization of nRNP Antigen in Mammalian Cells Stained with Peroxidase-Labelled IgG Fraction of Anti-nRNP Antibody Obtained from a Patient with Mixed Connective Tissue Disease (MCTD). Tohoku J. exp. Med., 1987, 153 (1), 21-26 - The peroxidase-labelled IgG fraction of anti-nRNP antibody obtained from a MCTD patient stained the nuclei of almost any kind of cells of man and rat. In addition, the cytoplasma of anterior horn cells of the rat spinal cord was exclusively reacted with this antibody. Transitory Type of Sphingomyelinase Deficient Niemann-Pick Disease: Clinical and Morphological Studies and Follow-Up of Two Sisters. Tohoku J. exp. Med., 1987, 153 (1), 27-36 - The transitory type is a rare and ill-defined variant of sphingomyelinase deficient NPD. Here reported are full clinical and morphological studies on two sisters with the transitory type. Our cases have the following features in common: 1) prominent visceral involvement from early infancy, such as marked hepatosplenomegaly, pulmonary infiltration, and numerous characteristic foam cells in the bone marrow aspirates; 2) very low sphingomyelinase activities in cultured skin fibroblasts; 3) atypical cherry-red spots in the retina; despite 4) absence of any neurological symptoms even at the age of 5 years and 6 months, and 3 years and 9 months, respectively. Furthermore, a few lines of evidence indicating nervous system involvement are also disclosed, that is, 5) vacuolated macrophages in the cerebrospinal fluid and ultrastructurally typical inclusion bodies in axons and Schwann cells in the rectal biopsy specimens, which were firstly revealed in this type of NPD. It is stressed that every case with apparent type B NPD should be searched for the presence or absence of neuronal involvement by not merely fundoscopy but other available means as we employed, which would greatly contribute to elucidate the clinico-pathogenetical knowledge not to mention the current classification of NPD.抄録全体を表示PDF形式でダウンロード (1497K) -

Med., 1987, 153 (1), 37-42 - A 70-year-old woman with Waldenström's macroglobulinemia (WMG) limited to mediastinal lymph nodes is described. The long-term course of 9 years was uneventful with the combination chemotherapy including cyclophosphamide, vincristine, 6-mercaptopurine and prednisolone. In WMG pleuropulmonary involvement is generally seen as a part of multisystemic disease. However, the cases showing pleuropulmonary manifestations as the sole initial clinical symptom have been rarely reported. In the present case WMG has been localized in the mediastinal lymph nodes during the period of 9 years. Med., 1987, 153 (1), 43-48 - Histopathological classification of gastric aberrant pancreas has been done by Heinrich's criteria. However in some case it was difficult to detect incomplete Langerhans' island consisting of several endocrine cells or a single endocrine cell by usual H-E staining. To identify the distribution of incomplete Langerhans' islands and endocrine cells, we applied an enzyme antibody method utilizing PAP method for 7 cases of gastric aberrant pancreas. It became easy to identify the endocrine cells such as single cell and islet cells by using the PAP method. In all 3 cases of aberrant pancreas classified as Heinrich type II and one of 2 case of type III, Langerhans' islands or endocrine small cell groups and single cells were found. Med., 1987, 153 (1), 49-54 - The association of multicentric giant lymph node hyperplasia (MGLH) of plasma cell type with monoclonal gammopathy was observed in a 37-year-old man. Tohoku J. exp. Med., 1987, 153 (1), 55-66 - The recently established culture medium, GIT, is applicable to many kinds of cells including mouse and human myeloma cells, and most adhesive cell lines. We applied this GIT medium to mouse B cell hybridoma production. When the medium was used to propagate myeloma cells before cell fusion and also for HAT selective medium, the fusion efficiency was more than twice as high as when the regular medium (RPMI-1640 supplemented with FBS) was used. Constantly more than 80% wells were hybridoma positive irrespective of the antigens used. To determine the optimal cell concentration at the hybridoma selection, a graded number of myeloma and spleen cells was distributed to each well; the best result was obtained when 3×104 myeloma and 3×105 spleen cells were distributed to each well. In addition, the GIT medium shows very little lot-to-lot variation. These results indicate that fusion efficiency in mouse B cell hybridoma production was greatly improved by using GIT medium.抄録全体を表示PDF形式でダウンロード (1115K) -

Med., 1987, 153 (1) 67-74 - The circadian rhythm of blood pressure (BP) was compared between patients with Cushing's syndrome and those with essential hypertension. In patients with essential hypertension, clear nocturnal falls in systolic and diastolic BP and heart rate (HR) were observed, and there was a positive correlation between HR and systolic or diastolic BP. On the other hand, in patients with Cushing's syndrome, there was no nocturnal fall in BP and instead a rise in some cases. In all cases with Cushing's syndrome there was a nocturnal fall in HR, and consequently no significant correlation between HR and BP in these patients. The present results imply that the normal circadian rhythm of blood pressure may be regulated at least in part by the hypothalamo-pituitary-adrenal system.抄録全体を表示PDF形式でダウンロード (663K) -

Med., 1987, 153 (1), 77-78 - Electrical stimulation of the presumptive pontine urine storage center, areas ventral to and/or lateral to the pontine micturition center, elicited an increase in threshold volume at which reflex micturition occurred in response to bladder filling with physiological saline, suggesting a facilitatory effect on the urine storage function. The electrolytic lesions made in the stimulated areas were histologically identified to distribute at the nucleus reticularis pontis oralis, Kölliker-Fuse nucleus and subcoeruleus.
